Exosome-loaded hydrogels: A new cell-free therapeutic approach for skin regeneration

Banafsheh Safari et al.

Summary: The treatment of unhealable and chronic cutaneous wounds is a significant challenge. Regenerative medicine based on mesenchymal stem cells (MSCs) has potential in skin repair. Exosomes, nano-sized vesicles with immune-tolerant properties, can easily transfer their content to recipient cells and play a role in wound healing through activating specific signaling pathways. To ensure the effectiveness of exosomes, the design of an optimized delivery system such as hydrogels is necessary.

Decellularized Tissues for Wound Healing: Towards Closing the Gap Between Scaffold Design and Effective Extracellular Matrix Remodeling

Victor Alfonso Solarte David et al.

Summary: The absence or damage of tissue is a major cause of diseases, and novel therapeutic alternatives aim to recover lost functions through tissue regeneration. Chronic cutaneous lesions are common wounds that require regenerative medicine and tissue engineering interventions to develop bioactive medical products for proper healing and prevention of complications. The coordination of cells, extracellular matrix (ECM), and biomolecules is crucial in tissue repair and regeneration, with ECM acting as a biological platform for their interactions. Tissue engineering utilizes synthetic polymers and decellularized tissues to mimic native ECM and provide necessary functionalities for cell differentiation.

Summary: ADSCs-EXOs modulate immune responses and inflammation, promote angiogenesis, accelerate proliferation and re-epithelization of skin cells, and regulate collagen remodelling to inhibit scar hyperplasia in wounds. Compared with ADSCs therapeutics, ADSCs-EXOs have high stability, easy storage, immune system non-rejection, homing effect, and easily controlled dosage. In patients with diabetes mellitus, ADSCs-EXOs can improve fat grafting and promote wound healing.
