Supramolecular Assembly of His-Tagged Fluorescent Protein Guests within Coiled-Coil Peptide Crystal Hosts: Three-Dimensional Ordering and Protein Thermal Stability

Ryan W. Curtis et al.

Summary: This study presents a method for including and overgrowing His-tagged fluorescent proteins within three-dimensional coiled-coil peptide crystals with metal ions. The fluorescent proteins are highly included within the crystal structure, displaying order and tight packing that enable energy transfer. This strategy may be useful for prolonged storage of thermally sensitive biopolymer guests within a 3D crystalline matrix.

Co-assembled Coiled-Coil Peptide Nanotubes with Enhanced Stability and Metal-Dependent Cargo Loading

Michael D. Jorgensen et al.

Summary: This study revealed the co-assembly of coil-coil peptides with metal ion ligands for hierarchical assembly into nanotubes with spatial control, enhancing stability in phosphate-buffered saline in a metal-dependent manner. Spatial control also allowed for site-specific labeling with His-tagged fluorophores through a metal-dependent manner.

A comparison of the collagen triple helix and coiled-coil peptide building blocks on metal ion-mediated supramolecular assembly

Ryan W. Curtis et al.

Summary: Metal ion-mediated assembly of peptides is a fascinating method for developing novel biomaterials. By functionalizing peptide building blocks with metal-binding ligands, hierarchical structures with diverse potential applications can be achieved. The identity and placement of the metal-binding ligands on the building blocks play crucial roles in the bottom-up development of the assemblies, leading to significant differences in structural morphologies.

Nanofibers Produced by Electrospinning of Ultrarigid Polymer Rods Made from Designed Peptide Bundlemers

Kyunghee Kim et al.

Summary: This study successfully demonstrates the assembly of peptide molecules into hierarchical nanostructured fibers, forming rigid-rod polymers with controlled structure and ultimately creating uniaxially oriented fibers. Characterization of the nanofibers using various microscopy and spectroscopy techniques reveals that they are composed of oriented rigid-rod chains with unique structural characteristics. The use of bundlemers chains for nanofiber fabrication shows promise for developing new fiber materials with targeted structure and properties.

Coiled coils 9-to-5: rational de novo design of α-helical barrels with tunable oligomeric states

William M. Dawson et al.

Summary: The rational design of linear peptides that assemble in water is challenging, as unique target structures must be encoded and alternative states avoided. Weak non-covalent forces that stabilize and discriminate between states present a challenge. However, progress has been made in rational de novo design for alpha-helical coiled-coil assemblies.
