Platelet ACKR3/CXCR7 favors antiplatelet lipids over an atherothrombotic lipidome and regulates thromboinflammation

Malgorzata Cebo et al.

Summary: Platelet ACKR3/CXCR7 surface expression is enhanced in coronary artery disease (CAD) patients, and it influences the prognosis. This study shows that ACKR3/CXCR7 regulates thromboinflammatory response by impacting the platelet lipidome. The findings suggest that ACKR3/CXCR7 may be a potential therapeutic target for cardiovascular diseases and CAD.

Community guidelines for GPCR ligand bias: IUPHAR review 32

Peter Kolb et al.

Summary: GPCRs regulate various physiological processes and their effects depend on the pairing of a receptor and a ligand. Ligands that induce biased signalling can lead to better drug effects and fewer side effects. However, ligand bias is complex, making it necessary to develop guidelines for designing and reporting biased signalling experiments.

ACKR3 regulates platelet activation and ischemia-reperfusion tissue injury

Anne-Katrin Rohlfing et al.

Summary: Platelet activation and thrombus formation play a critical role in tissue injury during ischemia/reperfusion. The absence of ACKR3 on platelets enhances platelet activation and thrombosis, exacerbating tissue damage. Activation of platelet-ACKR3 can inhibit platelet activation and thrombus formation, attenuating tissue injury.

Summary: The development and validation of the OPLS4 force field, which builds upon previous work with OPLS3e, improved model accuracy on challenging regimes of drug-like chemical space. A novel parametrization strategy for charged species was introduced, leading to improved accuracy on benchmarks assessing small-molecule solvation and protein-ligand binding.

Summary: The study compared the activation of ACKR3 by ADM and PAMP, finding that PAMP had a stronger potency towards ACKR3 than ADM. Importantly, PAMP-12 was efficiently internalized by ACKR3 without inducing G protein or ERK signaling in vitro.
