Identification of Liver Cancer Stem Cell Stemness Markers Using a Comparative Analysis of Public Data Sets

Kirill Borziak et al.

Summary: Analyzing single-cell transcriptomics data of liver cancer and liver progenitor cell types revealed elevated expression of stemness markers in CSCs compared to progenitor cell types, potentially explaining their differentiation and replication potential. These findings contribute to understanding the phenotypic profile of CSCs and important genes associated with them.

A genome-scale CRISPR screen reveals factors regulating Wnt-dependent renewal of mouse gastric epithelial cells

Kazuhiro Murakami et al.

Summary: Safe utilization of the regenerative potential of adult stem cells for clinical applications requires a comprehensive understanding of the regulatory mechanisms. Epithelial organoid cultures accurately mimic in vivo stem cell-driven epithelial renewal, offering a great ex vivo platform for studying key regulatory mechanisms. Through genome-scale CRISPR knockout screening, Alk, Bclaf3, and Prkra were identified as potential regulators of gastric epithelial differentiation and could suppress stemness/proliferation.

Glucocorticoid agonists enhance retinal stem cell self-renewal and proliferation

Kenneth N. Grise et al.

Summary: This study identified synthetic glucocorticoid agonists as regulators of retinal stem cell proliferation in culture, showing that they can increase RSC self-renewal and progenitor proliferation without influencing differentiation in vitro. Injection of these agonists into adult mouse eyes also induced proliferation in the ciliary epithelium, where adult RSCs reside, suggesting a potential activation of endogenous RSCs by glucocorticoids.

Cooperative genetic networks drive embryonic stem cell transition from naive to formative pluripotency

Andreas Lackner et al.

Summary: Genetic screens and CRISPR/Cas9 gene disruption were used to study the regulatory circuits governing the exit from naive pluripotency in mammalian embryos. The transcriptome profiles of ESCs deficient for regulators revealed delays in the transition from naive to formative state, with gene networks conserved in primate embryos. Integrated analysis of mutant transcriptomes identified regulatory modules and the role of signalling pathways in directing this cell state transition.

Transcriptional Profiling of Tumorspheres Reveals TRPM4 as a Novel Stemness Regulator in Breast Cancer

John Verigos et al.

Summary: Cancer stem cells play a significant role in chemoresistance, tumor recurrence, and metastasis in breast cancer, making them a promising target for new therapies. Through RNA-sequencing, researchers identified differential gene expression patterns between breast CSCs and non-CSCs, with the TRPM4 gene being found to be overexpressed in tumorspheres and affecting stemness properties. Inhibiting TRPM4 showed potential anti-tumor effects by targeting the bCSC subpopulation, suggesting a novel therapeutic approach for improving breast cancer treatments.

Novel induction of CD40 expression by tumor cells with RAS/RAF/PI3K pathway inhibition augments response to checkpoint blockade

Chi Yan et al.

Summary: The study identified that Rigosertib (RGS) can sensitize melanoma tumors to immune checkpoint blockade therapy by modulating immune cells in the tumor microenvironment, leading to tumor growth inhibition. Results suggest that RGS treatment induces immunogenic cell death in melanoma cells and upregulates CD40 expression, enhancing beneficial T-cell responses and improving patient survival. This indicates the potential therapeutic use of RGS in combination with immune checkpoint blockade for melanoma patients who do not respond to immune checkpoint blockade alone.

MYC dysfunction modulates stemness and tumorigenesis in breast cancer

Yiqiu Liu et al.

Summary: Our study using multiple databases found that MYC expression varies in different subtypes of breast cancer, particularly in TNBC. As a transcription factor, MYC not only participates in rewiring cancer signaling pathways like estrogen, WNT, NOTCH, but also interacts with cancer stem cells. This suggests that MYC plays an important role in breast cancer development and its amplification can serve as a predictor of diagnosis and prognosis.

Secretory Mucin SAC Promotes Neoplastic Progression by Augmenting KLF4-Mediated Pancreatic Cancer Cell Sternness

Koelina Ganguly et al.

Summary: Genetic ablation of secreted mucin 5AC (MUC5AC) in a murine model significantly delays neoplastic onset and PanIN lesion progression in pancreatic cancer, with a reduction in cancer stem cell markers and tumor formation frequency. Mechanistically, MUC5AC potentiates oncogenic signaling through integrin ow135, pSrc (Y416), and pSTAT3 (Y705), leading to enrichment of the self-renewing CSC population. These findings highlight the crucial role of MUC5AC in promoting cancer cell sternness during pancreatic tumorigenesis and suggest it as a potential therapeutic target.
