Investigation of the antileishmanial activity and mechanisms of action of acetyl-thiohydantoins

Bruna Taciane da Silva Bortoleti et al.

Summary: The study explored the antileishmanial effect of acetyl-thiohydantoins against Leishmania amazonensis, demonstrating inhibition of proliferation and alterations in parasite morphology and ultrastructure. The compounds induced an apoptosis-like process in the parasites by increasing ROS production and reducing TNF-alpha levels. Molecular docking analyses suggested potential pharmacophoric groups in the compounds that could be targeted for drug discovery in leishmaniasis treatment.

Tatiane Renata Fagundes et al.

Summary: This study evaluated the potential antitumor effect of thiohydantoin on MCF-7 breast cancer cells. The results showed that thiohydantoin treatment decreased cell viability and induced apoptosis and increased autophagy in the cells. This cytotoxic effect is linked to the increase in ROS production.

Thiohydantoins as anti-leishmanial agents: n vitro biological evaluation and multi-target investigation by molecular docking studies

Priscila Goes Camargo et al.

Summary: Leishmaniasis is a neglected tropical disease with limited treatment options. Thiohydantoins, a class of substances, show promising anti-leishmanial activity against the parasite in vitro and low cytotoxicity to human cells. These compounds induce oxidative stress and a late apoptosis-like process in the parasites. Molecular docking analysis suggests that the thiohydantoin ring may serve as a pharmacophoric group.
