Visualization of ligand-induced c-MYC duplex-quadruplex transition and direct exploration of the altered c-MYC DNA-protein interactions in cells

Summary: In this study, a new fluorescent probe ISCH-MYC was developed for specific recognition of c-MYC G-quadruplex based on the G-quadruplex-Triggered Fluorogenic Hybridization (GTFH) strategy. The probe allowed for the detection of duplex-quadruplex transition in c-MYC G-rich DNA in cells, and was used to study the interactions of transcription factors Sp1 and CNBP with c-MYC G-rich DNA in the presence of G-quadruplex stabilizing ligand PDS. This research provides a visualization strategy for exploring the mechanism of G-quadruplex stabilizing ligands on c-MYC G-rich DNA and relevant proteins.

DNA methylome profiling of circulating tumor cells in lung cancer at single base-pair resolution

Summary: In this study, an efficient workflow LCM-mu WGBS was introduced to profile the DNA methylome of circulating tumor cells from lung cancer patients. A unique DNA methylation signature of CTCs distinct from primary lung cancer tissues was identified, with promoter hypermethylation of epithelial genes as a hallmark of stable epithelial-mesenchymal transition. Furthermore, CTCs may exhibit a stemness-related feature during dissemination and metastasis, providing potential noninvasive CTC DNA methylation biomarkers for clinical diagnosis.
