The role of sentinel lymph node mapping in colon cancer: detection of micro-metastasis, effect on survival, and driver of a paradigm shift in extent of colon resection

Sukamal Saha et al.

Summary: Sentinel lymph node mapping in colon cancer is primarily used for accurate nodal staging and to guide limited colon resection in early T stage tumors. The incidence of micro-metastasis was reviewed in this study, with positive results indicating that SLNM results in more positive nodes per patient and longer survival rates when treated with chemotherapy.

CXCR4/ACKR3/CXCL12 axis in the lymphatic metastasis of vulvar squamous cell carcinoma

Natalia Rusetska et al.

Summary: The study found that the CXCR4/ACKR3/CXCL12 axis is activated during progression and lymphatic spread of VSCC. CXCR4 and ACKR3 proteins are nearly absent in precancerous lesions but are strong in VSCC samples. Additionally, serum CXCL12 levels are significantly increased in both patients with VSCC and premalignant vulvar lesions.

Lymph node colonization induces tumor-immune tolerance to promote distant metastasis

Nathan E. Reticker-Flynn et al.

Summary: Lymph node metastasis plays an active role in shaping distant metastasis in solid malignancies, and this mechanism involves the evasion of immune cells, colonization in lymph nodes, and subsequent colonization in other tissues. This mechanism is conserved in human cancers and murine cancer models.

CD4+ and CD8+ T cells in sentinel nodes exhibit distinct pattern of PD-1, CD69, and HLA-DR expression compared to tumor tissue in oral squamous cell carcinoma

Krzysztof Piersiala et al.

Summary: This study investigated the expression of PD-1, CD69, and HLA-DR on T cells in OSCC patients' PBMCs, TDLNs, and tumor samples. The results showed highly activated T lymphocytes expressing high levels of PD-1 in OSCC tissue, with lower levels in TDLNs and peripheral blood. Patients with a high percentage of CD3(+) PD-1(+) cells in TDLNs had significantly lower disease-free and overall survival rates. This suggests that analyzing PD-1 levels in TDLNs may help predict survival and response to anti-PD-1 therapy in OSCC.

uPAR-expressing melanoma exosomes promote angiogenesis by VE-Cadherin, EGFR and uPAR overexpression and rise of ERK1,2 signaling in endothelial cells

Alessio Biagioni et al.

Summary: This study demonstrates the crucial role of uPAR in the pro-angiogenic activity of melanoma Exos, with uPAR knockout reducing the angiogenic effects of Exos in vitro and in vivo. Inhibition of signaling pathways in endothelial cells, uPAR-EGFR interaction, and EGFR phosphorylation can all contribute to the reduction of melanoma Exos-induced angiogenesis. The identification of uPAR-expressing Exos as a potential biomarker for evaluating angiogenic propensity and treatment response in metastatic melanoma patients is suggested.

Tenascin-C expression in the lymph node pre-metastatic niche in muscle-invasive bladder cancer

Christopher R. Silvers et al.

Summary: Elevated expression of tenascin-C in regional lymph nodes may be a predictor of bladder cancer metastasis and associated with worse survival. Urinary extracellular vesicles contain cytokines that are positively correlated with lymph node tenascin-C expression. Bladder cancer extracellular vesicles induce tenascin-C expression in fibroblasts in an NF-kappa B-dependent manner.

Postoperative Management of Salivary Gland Tumors

Nikhil P. Joshi et al.

Summary: Salivary gland tumors are a diverse group of neoplasms with varying histologies and outcomes. Treatment usually involves surgery for primary and gross nodal disease, with the management of clinically node-negative neck depending on lymph nodal risk. Both surgery and radiation can be used to address at-risk lymph nodes, especially for minor salivary gland tumors. Radiation plays a crucial role in reducing locoregional recurrence risk and controlling disease at the skull base.

Lymphangiogenesis-inducing vaccines elicit potent and long-lasting T cell immunity against melanomas

Maria Stella Sasso et al.

Summary: The induction of lymphatic growth in melanoma has been linked to poor prognosis, but recent research has shown that it can enhance cancer immunotherapy and boost T cell immunity. Developing a translational approach, the study found that inducing lymphangiogenesis in cancer vaccines can lead to stronger T cell activation and higher frequencies of antigen-specific T cells, ultimately providing effective tumor control and long-term immunological memory in mouse melanoma models.
