Electrochemical One-Step Immunoassay Based on Switching Peptides and Pyrolyzed Carbon Electrodes

Jun-Hee Park et al.

Summary: Switching peptides were designed as reversible binding antibodies for different antigens and source animals. An electrochemical one-step immunoassay using switching peptides labeled with ferrocene was conducted for quantitative measurement of analytes. The feasibility of this method was assessed by analyzing electrochemical properties and the factors affecting the amperometry of switching peptides labeled with ferrocene. The applicability of the electrochemical immunoassay for medical diagnosis of human hepatitis B surface antigen was evaluated.

Switching-peptides for one-step immunoassay and its application to the diagnosis of human hepatitis B

Ji-Hong Bong et al.

Summary: In this study, switching-peptides were used to develop a one-step immunoassay for detecting antigen-antibody interactions without the need for additional antibody treatment or washing steps. The design principles of switching-peptides and the study of their interaction with antibodies using thermodynamic parameters demonstrated successful binding properties. The feasibility of using switching-peptides for quantitative one-step immunoassay was confirmed through HBsAg detection and statistical comparison with conventional ELISA results, showing promising results for practical applications.

An NIR dual-emitting/absorbing inorganic compact pair: A self-calibrating LRET system for homogeneous virus detection

Dongkyu Kang et al.

Summary: A near-infrared self-calibrating luminescence resonance energy transfer system was developed for background-free detection in homogeneous sandwich-immunoassays. The inorganic LRET pair of NIR dual-emitting lanthanide-doped nanoparticles and NIR-absorbing lanthanide-doped nanoparticles showed stable detection of AIV nucleoproteins with a 0.38 pM limit of detection.

Microbial biosensor for Salmonella using anti-bacterial antibodies isolated from human serum

Jun-Hee Park et al.

Summary: This study presents a novel microbial biosensor for Salmonella detection based on impedance spectrometry using antibodies isolated from human serum. The isolated anti-Salmonella antibodies showed high binding affinity for S. enteritidis compared to other Gram-negative bacterial strains. The detection limit of the isolated antibodies was estimated to be 1.0 x 10(3) cells/mL for S. enteritidis and 1.0 x 10(6) cells/mL for BL21 (DE3).

Jaeyong Jung et al.

Summary: The study purified antibodies against SARS-CoV-2 NP from pig serum in two steps, demonstrating enhanced specificity through immunoassays and estimating binding constants using a biosensor. Competitive assays showed the potential of these antibodies to detect SARS-CoV-2 and differentiate it from other related coronaviruses in viral fluids.

Isolation of Antibodies Against the Spike Protein of SARS-CoV from Pig Serum for Competitive Immunoassay

Jaeyong Jung et al.

Summary: Several endemic corona viruses (eCoVs) are common causes of seasonal common cold and pneumonia, sharing sequence homology with SARS-CoV-2. Pig serum was used to isolate IgG antibodies against the spike protein of SARS-CoV, showing selectivity to viral antigens and a binding constant of approximately 1.5 x 10(-8) M. The isolated antibodies were successfully applied in immunoassays for SARS-CoV viral culture fluid, demonstrating feasibility for detection and quantification.

Rapid Analysis of Bacterial Contamination in Platelets without Pre-Enrichment Using Pig Serum-Derived Antibodies

Ji-Hong Bong et al.

Summary: This study presents a rapid analysis method for detecting bacterial contamination in platelet samples using antibodies derived from pig serum, without the need for pre-enrichment. The feasibility of using these antibodies against Gram-negative and Gram-positive bacteria was successfully demonstrated, and an impedance immunosensor for detecting contamination of platelets was developed.
