Podocalyxin molecular characteristics and endometrial expression: high conservation between humans and macaques but divergence in mice†

Bothidah Thach et al.

Summary: Podocalyxin is a key negative regulator of human endometrial receptivity and inhibits embryo implantation in mice. This study compared the expression patterns of Podocalyxin in human, rhesus macaque, and mouse uterus, and found that it is down-regulated in the luminal epithelium in all three species during receptivity. The molecular characteristics and regulation of Podocalyxin are more similar between humans and macaques, suggesting that macaques are a better animal model for studying the function of Podocalyxin in human fertility.

Podocalyxin promotes an impermeable epithelium and inhibits pro-implantation factors to negatively regulate endometrial receptivity

Sophea Heng et al.

Summary: PCX is a key negative regulator of human endometrial receptivity, suppressing genes controlling cell adhesion and communication while increasing expression of proteins governing epithelial barrier functions. It also suppresses receptivity-supporting factors and signaling pathways, and stimulates anti-implantation genes. Overall, PCX promotes an anti-adhesive and impermeable epithelium while negatively controlling endometrial receptivity for implantation.

Podocalyxin is a key negative regulator of human endometrial epithelial receptivity for embryo implantation

Sarah G. Paule et al.

Summary: This study identified PODX as a key negative regulator of endometrial receptivity, with its specific downregulation in the mid-secretory phase likely mediated by progesterone. Overexpression of PODX inhibited human embryo attachment and invasion in functional models, highlighting its role as a negative regulator of epithelial receptivity for implantation.

Podocalyxin inhibits human embryo implantation in vitro and luminal podocalyxin in putative receptive endometrium is associated with implantation failure in fertility treatment

Sophea Heng et al.

Summary: This study found that PCX inhibits embryo implantation in vitro and in IVF patients. High levels of PCX significantly inhibited blastocyst attachment and penetration. The presence of PCX in the luminal epithelium was negatively associated with the success rate of clinical pregnancy.
