Histone H2A variants: Diversifying chromatin to ensure genome integrity

Philipp Oberdoerffer et al.

Summary: Histone variants, such as H2A.X, H2A.Z, and macroH2A, play crucial roles in protecting the integrity of the genome through DNA damage response pathways. Their deposition, posttranslational modifications, and interactions with other proteins safeguard DNA from both endogenous and exogenous threats. Understanding the involvement of H2A variants in maintaining genome stability and their contributions to human diseases, including cancer, provides valuable insights.

Effects of soil-extractable metals Cd and Ni from an e-waste dismantling site on human colonic epithelial cells Caco-2: Mechanisms and implications

Jiao-Yang Ma et al.

Summary: This study assessed the toxic effects and underlying mechanisms of toxic metals released from e-wastes on human colonic epithelial cells. The findings revealed that the soil extracts induced cytotoxicity, oxidative damage, inflammatory response, and cell apoptosis. These results suggest that soil ingestion from e-waste dismantling sites may adversely impact human health.

Dermal uptake of chlorinated organophosphate flame retardants via contact with furniture fabrics; implications for human exposure

Mohamed Abou-Elwafa Abdallah et al.

Summary: The chlorinated organophosphate flame retardants (Cl-PFRs) have been widely used in upholstered furniture despite their carcinogenic potential. This study assessed the dermal absorption of these chemicals from furniture fabrics for the first time, revealing that all three target Cl-PFRs were dermally bioavailable to varying degrees. Factors such as compound properties, exposure matrix, exposure dose, and skin hydration can influence the dermal uptake of Cl-PFRs.

Polybrominated diphenyl ether quinone exposure leads to ROS-driven lysosomal damage, mitochondrial dysfunction and NLRP3 inflammasome activation

Bingwei Yang et al.

Summary: Exposure to PBDEs metabolite PBDEQ induces ROS and mitochondrial dysfunction, promoting NLRP3 inflammasome activation through the TLRs-NF-kappa B signaling pathway. Additionally, bromine metabolism and K+ efflux are involved in the activation of this inflammatory response.

BDE-209 and DBDPE induce male reproductive toxicity through telomere-related cell senescence and apoptosis in SD rat

Xiangyang Li et al.

Summary: This study found that both BDE-209 and DBDPE can cause testicular damage, decrease sperm quantity and motility, increase sperm malformation rates, and induce cell senescence and apoptosis in rat testis by shortening telomere length and reducing telomerase activity. BDE-209 had more severe effects on male reproduction compared to DBDPE.

Recent advances in γH2AX biomarker-based genotoxicity assays: A marker of DNA damage and repair

Nazanin Rahmanian et al.

Summary: The phosphorylation of histone variant H2AX and the formation of gamma H2AX are primary responses to DNA double-strand breaks, serving as robust and sensitive tools for diagnosing DNA damage and repair in pre-clinical drug discovery investigations. Gamma H2AX can also serve as a surrogate marker for drug-induced cytotoxicity, while recent advances in genomic research allow for its detection as a specific biomarker for quantitatively analyzing DNA damages and repair. The use of gamma H2AX as a biomarker shows promise in genotoxicity analyses and the monitoring and management of cancer diseases.

Mechanisms and Regulation of Cellular Senescence

Laureline Roger et al.

Summary: Cellular senescence is an essentially irreversible proliferative arrest state where cells release pro-inflammatory and proteolytic factors. Different types of senescent cells accumulate in various tissues and organs with unique functions. Understanding how cells undergo extensive changes to induce a common cellular state is crucial for cancer prevention and improving health during aging.

Inhibition of the P53/P21 Pathway Attenuates the Effects of Senescent Nucleus Pulposus Cell-Derived Exosomes on the Senescence of Nucleus Pulposus Cells

Chang-chun Chen et al.

Summary: The study found that SNPC-Exo treatment can induce NPC senescence, while inhibition of the P53/P21 pathway can attenuate SNPC-Exo-induced NPC senescence. Inhibition of the P53/P21 pathway led to an increase in NPC proliferation rate, a decrease in the percentage of SA-beta-gal-positive cells, and facilitated NPC entry into the S phase.

Environmentally-Induced (Extrinsic) Skin Aging: Exposomal Factors and Underlying Mechanisms

Jean Krutmann et al.

Summary: The skin, as a barrier organ, is an ideal model for studying environmentally-induced aging. This review explains how exposure to specific exposomal factors can lead to extrinsic skin aging, involving interactions between these factors and genetic modifications, ultimately causing macromolecular damage and changes in fibroblasts in the dermal compartment of the skin.

Mechanisms of Cellular Senescence: Cell Cycle Arrest and Senescence Associated Secretory Phenotype

Ruchi Kumari et al.

Summary: Cellular senescence is a stable cell cycle arrest triggered in response to various stimuli, characterized by dynamic changes in senescent cells. It has both positive effects on tissue repair and anti-cancer mechanisms, but can also negatively impact organismal health. Multiple pathways, including p53/p21 and p16/pRB, play central roles in regulating this process.

Polybrominated diphenyl ethers quinone exhibits neurotoxicity by inducing DNA damage, cell cycle arrest, apoptosis and p53-driven adaptive response in microglia BV2 cells

Zixuan Liu et al.

Summary: The study demonstrated that PBDEQ exposure could induce DNA damage, cell apoptosis, and G2/M phase cell arrest in a p53-dependent manner. Activation of p53 coordinated cell cycle progression, alleviated DNA damage, and ultimately mitigated apoptosis in BV2 cells. Antioxidant NAC inhibited p53 activation upon PBDEQ exposure, suggesting that PBDEQ-induced DNA damage and p53 activation were mediated by reactive oxygen species (ROS).

Organophosphorus flame retardants in breast milk from Beijing, China: Occurrence, nursing infant's exposure and risk assessment

Xuelei Chen et al.

Summary: The study investigated the occurrence of thirteen OPFRs in breast milk samples collected from Beijing mothers in 2018, revealing China as one of the most affected regions by OPFRs. Personal characteristics of mothers were found to impact the levels of certain OPFRs in breast milk, with most nursing infants' daily OPFR intake not raising significant health concerns. However, two OPFRs, TPhP and EHDPP, were noted to have higher estimated daily intakes than corresponding reference doses, potentially posing health risks.
