In Situ Detection of Nanotoxicity in Living Cells Based on Multiple miRNAs Probed by a Peptide Functionalized Nanoprobe

Di Han et al.

Summary: In this study, a strategy was developed for sensitive detection of nanotoxicity using a functional nanoprobe loaded with molecular beacons. The nanoprobe efficiently delivered molecular beacons into living cells to monitor miRNA changes and evaluate nanotoxicity. The results showed that this nanoprobe is more sensitive compared to conventional methods for studying cytotoxicity.

Dispersion-to-localization of catalytic hairpin assembly for sensitive sensing and imaging microRNAs in living cells from whole blood

Huijie Bai et al.

Summary: A novel strategy, DL-CHA, has been developed for sensitive sensing and imaging of miRNAs in living cells. This approach reduces sequence design complexity by using only three classes of DNA strands and significantly improves the S/B ratio. The study provides a powerful tool for intracellular molecules detection and broadens the design space for constructing high-performance DNA circuits.

DNA Nanowire Guided-Catalyzed Hairpin Assembly Nanoprobe for In Situ Profiling of Circulating Extracellular Vesicle-Associated MicroRNAs

Ye Zhang et al.

Summary: Extracellular vesicle-associated miRNAs (EV-miRNAs) provide a new noninvasive biomarker for disease diagnosis. The DNAnanowire guided-catalyzed hairpin assembly (NgCHA) nanoprobe is designed for accurate profiling of EV-miRNAs. The platform shows high sensitivity and can be used for early stage breast cancer diagnosis and risk assessment.

Flap Endonuclease 1-Assisted DNA Walkers for Sensitively and Specifically Sensing ctDNAs

Xianyi Cheng et al.

Summary: A DNA walker assisted by flap endonuclease 1 (FEN 1) has been proposed to achieve mutant biosensing with high specificity and sensitivity. The system is capable of discriminating single-base mutant DNA targets from wild-type DNA, and can detect DNA targets as low as 1 fM and mutation abundance as low as 0.1%. Detection of epidermal growth factor receptor (EGFR) L858R mutation on cell-free DNA samples showed results consistent with next-generation sequencing, indicating the potential of the DNA walker for liquid biopsy.
