Two Faces of Neutrophils in Liver Disease Development and Progression

Yeonhee Cho et al.

Summary: Neutrophils, the most abundant type of leukocyte in human blood, play a major role in host defense against invading pathogens and in sterile injury. However, under chronic inflammatory conditions, neutrophils may lose their self-resolving capabilities and maintain an inflammatory phenotype, exacerbating tissue damage. This review aims to provide a better understanding of how neutrophils interact with tissue microenvironments and acquire disease-specific phenotypes in various liver diseases.

Myeloid-Cell-Specific IL-6 Signaling Promotes MicroRNA-223-Enriched Exosome Production to Attenuate NAFLD-Associated Fibrosis

Xin Hou et al.

Summary: This study investigated the role of IL-6 signaling in NAFLD by regulating antifibrotic microRNA-223 in myeloid cells. The results showed that myeloid-specific IL-6 signaling inhibits liver fibrosis through exosomal transfer of antifibrotic miR-223 into hepatocytes. These findings provide potential therapeutic targets for the treatment of NAFLD.

Neutrophil-to-hepatocyte communication via LDLR-dependent miR-223-enriched extracellular vesicle transfer ameliorates nonalcoholic steatohepatitis

Yong He et al.

Summary: This study reveals that the elevation of miR-223 in hepatocytes is due to preferential uptake of miR-223-enriched EVs released from neutrophils and other cells, mediated by LDLR and APOE expression. The internalized EV-derived miR-223 acts to inhibit hepatic inflammatory and fibrogenic gene expression, thereby limiting the progression of NASH.

Alcohol-associated liver disease in the United States is associated with severe forms of disease among young, females and Hispanics

Ashwani K. Singal et al.

Summary: Through analysis of data from the NHANES, NIS, and UNOS databases, it was found that young age, female gender, and Hispanic race are independently associated with ALD-related burden and acute on chronic liver failure (ACLF) risk in the United States.

Diagnosis and Treatment of Alcohol-Associated Liver Disease A Review

Ashwani K. Singal et al.

Summary: Alcohol-associated liver disease is a major cause of cirrhosis, with significant risk factors including amount and duration of alcohol consumption, gender, obesity, and specific genetic polymorphisms. Abstinence can greatly improve survival rates for patients with alcohol-associated liver disease.

Early liver transplantation for acute alcoholic hepatitis: We can't say no

Philippe Mathurin

Summary: For severe alcoholic hepatitis patients not responding to medical therapy, postponing the decision to list for transplantation is harmful as there are no other therapeutic alternatives available. Early transplantation, with a strict selection process, has gained increasing support from experts globally, but there is still significant heterogeneity in its application across countries and even within the same country.

Super enhancer regulation of cytokine-induced chemokine production in alcoholic hepatitis

Mengfei Liu et al.

Summary: Alcoholic hepatitis is characterized by intense liver inflammation driven by excessive cytokines and chemokines production and immune cell infiltration. The study identified a super-enhancer that regulates the expression of multiple CXCL chemokines in alcoholic hepatitis and may be a potential therapeutic target.

Bile acid-activated macrophages promote biliary epithelial cell proliferation through integrin αvβ6 upregulation following liver injury

Adrien Guillot et al.

Summary: The study reveals that BEC injury leads to processes including cholestasis, recruitment of CCR2(+) monocytes, and BEC proliferation, ultimately inducing the expression of integrin ITGβ6 to promote BEC proliferation, potentially serving as therapeutic targets for cholangiopathies.

MicroRNAs as regulators, biomarkers and therapeutic targets in liver diseases

Xiaolin Wang et al.

Summary: MicroRNAs are small, non-coding RNAs that play crucial roles in regulating liver functions and are associated with various liver diseases, making them promising for early diagnosis and treatment strategies. Continued research on miRNAs in the liver may lead to the identification of biomarkers and therapeutic targets for liver diseases in the future.

Neutrophils in liver diseases: pathogenesis and therapeutic targets

Kai Liu et al.

Summary: Previously considered a homogeneous population, recent data have shown that neutrophils are diverse and involved in tissue damage and immune regulation. Their phenotypic and functional plasticity has been identified in various diseases, including cancer, inflammatory disorders, and infections. Neutrophils play a role in liver diseases through their autocrine, paracrine, and immune modulation functions.
