Understanding the mechanisms of these ncRNAs can lead to the development of novel therapeutic approaches targeting CIN in cancer patients.

Paradoxes of cancer: Survival at the brink

Summary: Our understanding of cancer initiation, persistence, and progression is evolving, but traditional somatic mutation theory alone cannot explain cancer's resistance to extinction. We propose a more systems-based framework that focuses on gene regulatory networks and cell-fate decisions to better understand cancer complexity. By exploring the adaptive exploration of these networks at the edge of order and chaos, we can explain cancer paradoxes and the role of polyploidy in accessing the oncofetal state. We challenge researchers to consider cancer defects as adaptation tools that have arisen during evolution and are intrinsic to cancer, which could lead to more successful anti-cancer treatments.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Summary: Lamins are components of the nuclear lamina and play a role in transcriptional regulation and chromatin organization. Recent studies have shown that lamins not only regulate gene transcription in regions associated with the nuclear lamina, but also interact with promoters and enhancers located inside the nucleus. Functional studies suggest that lamins play a role in transcription and chromatin interactions of key developmental gene programs. This review focuses on the emerging non-canonical functions of lamins in Drosophila and mammals as model organisms.

Systemic delivery of antisense oligonucleotides rescues tissue histophysiology and increases the lifespan of a progeria mouse model.

Embryonic Programs in Cancer and Metastasis-Insights From the Mammary Gland

Summary: Cancer is a reversion of differentiated cells to a primitive state resembling embryonic cells. This review examines the developmental mechanisms involved in embryonic mouse mammary gland development and their roles in mammary gland tumorigenesis and metastasis. The study reveals the reactivation or dysregulation of key embryonic developmental pathways in cancer, leading to abnormal proliferation, epithelial-to-mesenchymal transition, and altered stem cell potency. Recent studies using lineage tracing and transcriptomics have identified features of embryonic cells in cancer and metastasis. Embryonic molecular signatures may serve as prognostic markers and contribute to the discovery of novel therapeutic targets and strategies.

The co-evolution of the genome and epigenome in colorectal cancer
