Cell cycle regulation: p53-p21-RB signaling

Kurt Engeland

Summary: RB and p53 are central tumor suppressors that play key roles in regulating the cell division cycle. RB downregulates numerous genes by forming complexes with E2F transcription factors, and its repression is released through phosphorylation. p53 activates CDKN1A gene to regulate RB phosphorylation, leading to expression of cell cycle regulators. There is an overlap between RB and DREAM signaling pathways.

In vitro anticancer effects of alpelisib against PIK3CA-mutated canine hemangiosarcoma cell lines

Marika Maeda et al.

Summary: Hemangiosarcoma (HSA) is a malignant neoplasm with poor prognosis, especially in canines, and can serve as a valuable model for intractable human diseases. The high frequency of PIK3CA mutations in canine HSA affects the EGFR signaling pathway, and the molecular targeted drug alpelisib shows significant antitumor effects against these mutations.

The Mechanistic Understanding of RAD51 Defibrillation: A Critical Step in BRCA2-Mediated DNA Repair by Homologous Recombination

Fabrizio Schipani et al.

Summary: The cytotoxic action of anticancer drugs can be enhanced by inhibiting DNA repair mechanisms, and the fibrillation of RAD51 plays a crucial role in homologous recombination and DNA repair. This study discovered that BRC4 can reverse RAD51 fibrillation, prevent its nuclear transport, and inhibit homologous recombination to enhance the efficacy of chemotherapy treatment.

Inhibiting homologous recombination by targeting RAD51 protein

A. Demeyer et al.

Summary: Homologous recombination (HR) plays a crucial role in repairing DNA double-strand breaks and regulating HR is essential for maintaining genomic stability. Over-activation of HR in many forms of cancer leads to increased tumor resistance to DNA-damaging treatments. RAD51, the core protein of HR, is often over-expressed in cancers and targeting RAD51 directly can sensitize cancer cells to existing therapies.

From Conventional to Precision Therapy in Canine Mammary Cancer: A Comprehensive Review

Guillermo Valdivia et al.

Summary: Canine mammary tumors are common in intact female dogs, with mammary cancer representing a significant portion. Studies on new treatments for canine mammary cancer are promising and can provide insights into human breast cancer research. The lack of translation from in vitro studies to clinical trials in dogs with CMC is highlighted as a gap in current research.

Overexpression of HMGA1 confers radioresistance by transactivating RAD51 in cholangiocarcinoma

Jianping Song et al.

Summary: This study demonstrated that overexpression of high mobility group A1 (HMGA1) in cholangiocarcinomas is linked to poor prognosis and promotes cell proliferation and invasion. HMGA1 also enhances radioresistance by transcriptionally activating RAD51, a process that can be reversed by inhibiting RAD51.
