Benchmarking the ideal sample thickness in cryo-EM

Summary: The study analyzed the relationship between sample thickness and data quality using MicroED, revealing that structures can be reliably determined from crystalline lamellae up to twice the inelastic mean free path. Lower resolution diffraction was observed at three times the mean free path for all three accelerating voltages, but insufficient data quality prevented structure determination. No coherent diffraction was observed from lamellae thicker than four times the calculated inelastic mean free path, setting an ideal specimen thickness benchmark for cryo-EM methods.

Cryo-EM single-particle structure refinement and map calculation using Servalcat

Summary: In 2020, cryo-EM single-particle analysis achieved true atomic resolution with the increase in high-resolution reconstructions. The new Python package and program called Servalcat is designed to facilitate atomic model refinement through a refinement pipeline using REFMAC5 program, calculating difference maps and enhancing weak features. Servalcat also supports atomic model refinement under symmetry constraints for asymmetric unit models.

MyD88 TIR domain higher-order assembly interactions revealed by microcrystal electron diffraction and serial femtosecond crystallography

Summary: MyD88 and MAL are TLR adaptors that induce pro-inflammatory cytokine production, with the TIR domain of MAL forming filaments in vitro that nucleate the assembly of crystalline arrays of the MyD88 TIR domain. The structures of these assemblies were determined using MicroED and SFX techniques, showing the importance of the MyD88 interface residues for TLR4 signaling in vivo. The study provides insight into TLR signal transduction and compares the MicroED and SFX techniques.
