Dietary bovine milk miRNAs transported in extracellular vesicles are partially stable during GI digestion, are bioavailable and reach target tissues but need a minimum dose to impact on gene expression

Maria-Carmen Lopez Hazas et al.

Summary: Extracellular RNAs are unstable and can be degraded, but milk EVs protect and promote the uptake of miRNAs. However, the concentration of miRNAs in EVs may not be sufficient for gene modulation, although orally delivered EV-loaded miRNAs can reach tissues in vivo, potentially exerting biological effects. Further research is warranted for potential implications in nutrition and health.

A high-concentrate diet induces an inflammatory response and oxidative stress and depresses milk fat synthesis in the mammary gland of dairy cows

N. Ma et al.

Summary: Feeding a high-concentrate diet to dairy cows can lead to metabolic diseases, such as subacute ruminal acidosis (SARA) and milk fat depression. This study aimed to investigate the effects of a high-concentrate diet on the inflammatory response, oxidative stress, and milk fat synthesis in the mammary gland of dairy cows. The results showed that a high-concentrate diet induced SARA, increased the concentration of lipopolysaccharide (LPS) in the peripheral blood, stimulated inflammation and oxidative stress, and inhibited milk fat synthesis in the mammary gland.

MiRNAs in milk can be used towards early prediction of mammary gland inflammation in cattle

T. Tzelos et al.

Summary: This study examines the potential of miRNAs as early diagnostic biomarkers for bovine mastitis. The levels of four inflammation-associated miRNAs were found to be significantly correlated with mRNA levels of known inflammatory markers in milk cells. The analysis also revealed that miRNA levels increase with the severity of mammary inflammation and decrease in later lactations. The study concludes that miRNA levels can accurately identify early inflammatory changes in individual milk samples.

Bovine Milk-Derived Exosomes as a Drug Delivery Vehicle for miRNA-Based Therapy

Lorena del Pozo-Acebo et al.

Summary: miRNAs are potential mediators of gene expression in therapeutic strategies, but the instability of extracellular RNAs limits their biological function. Strategies to enhance miRNA therapeutic potential include developing efficient, tissue-specific delivery methods using exosomes. However, a standard, reproducible, and cost-effective protocol for exosome isolation and extracellular RNA delivery is currently lacking.

Huaiyu Mi et al.

Summary: PANTHER is a resource for evolutionary and functional classification of protein-coding genes, with extensive improvements made in the past two years, including the refactoring of Protein Class ontology and addition of services for programmatic access. The resource provides comprehensive classification and annotation information for genes from different taxonomic groups and also allows users to add their own protein sequences for evolutionary context and fine-grained annotations.

Abundance of solute carrier family 27 member 6 (SLC27A6) in the bovine mammary gland alters fatty acid metabolism

Huimin Zhang et al.

Summary: Milk fatty acid composition varies with lactation stages, with significant changes in concentrations of different fatty acids. Transcriptome profiling revealed key genes and pathways involved in lipid metabolism, with an emphasis on FA transport mechanisms and their impact on milk FA composition. Silencing of SLC27A6 in mammary cells altered gene expression associated with FA activation, oxidation, and transport, leading to changes in the intracellular content of triglycerides and specific FA profiles. These findings suggest a central role of SLC27A6 in regulating FA metabolism in bovine mammary epithelial cells.
