Photobiocatalytic Oxyfunctionalization with High Reaction Rate using a Baeyer-Villiger Monooxygenase from Burkholderia xenovorans in Metabolically Engineered Cyanobacteria

Elif Erdem et al.

Summary: BVMOXeno is a highly efficient Baeyer-Villiger monooxygenase with higher reaction rates and lower KM value compared to currently known BVMOs, almost completely suppressing unwanted side reactions. The recombinant production of BVMO in cyanobacteria allows more efficient utilization of water and oxygen generated by photosynthesis.

High efficient production of plant flavonoids by microbial cell factories: Challenges and opportunities

Jiachen Sun et al.

Summary: Plant flavonoids are secondary metabolites with various physiological and pharmacological activities. Current production methods from plants or chemicals do not meet the requirements of green and sustainable development. Microbial synthesis of flavonoids has shown potential for large-scale production, but challenges such as metabolic flux imbalance and conflict between cell growth and production hinder commercialization of microbial cell factories. Strategies discussed in this article, combined with protein structure prediction, de novo protein design, artificial intelligence, biocatalytic retrosynthesis, and intelligent stress resistance, provide insights for efficient production of plant flavonoids and other natural products in microbial cell factories.

Carbon-negative synthetic biology: challenges and emerging trends of cyanobacterial technology

Chunlin Tan et al.

Summary: Global warming and climate instability have sparked interest in using renewable carbon resources for sustainable chemical production. Cyanobacteria, with their ability to directly use light and CO2 as energy and carbon sources, respectively, are ideal cellular factories for carbon-negative production of chemicals. However, adapting cyanobacterial technology to industry still faces challenges such as low productivity, poor tolerance, and difficulties in product harvesting, which may be addressed by synthetic biology.

Alginate immobilization as a strategy for improving succinate production during autofermentation using cyanobacteria Synechocystis sp. PCC 6803

Daniel A. Alfaro-Sayes et al.

Summary: This study investigated the immobilization of a cyanobacterial strain for succinate production. The results showed that immobilized cells had higher succinate yield and dry cell weight compared to free cells during continuous autofermentation. In addition, the immobilization process allowed for shorter cultivation time. Continuous batch culture proved to be a convenient strategy for utilizing immobilized cyanobacteria, resulting in a simpler and faster process with increased succinate production rate in each cycle.

ATP and NADPH engineering of Escherichia coli to improve the production of 4-hydroxyphenylacetic acid using CRISPRi

Yu-Ping Shen et al.

Summary: This study developed a novel strategy called CECRiS for improving NADPH and/or ATP availability, enhancing the production of 4HPAA in engineered E. coli. Through CRISPRi screening, genes consuming NADPH and ATP were identified and repressed, leading to the highest yield and titer of 4HPAA to date.

Eleftheria-Angeliki Valsami et al.

Summary: This study investigated various cultivation conditions for heterologous beta-phellandrene production by Synechocystis sp. PCC 6803, finding that immobilized cells in calcium alginate beads resulted in higher production compared to suspended cultures. Additionally, extreme conditions of pH 10.5 and 600 mM NaCl proved to be conducive for cell growth and beta-phellandrene production, offering new approaches for sustainable production of terpenoids.
