Photoinduced Three-Component Difluoroamidosulfonylation/Bicyclization: Regioselectivity Synthesis of Seven-Membered Dibenzosultams

Summary: A mild and safe photochemical methodology has been developed for the synthesis of difluoroamidosulfonylated dioxodibenzothiazepines via a three-component radical bicyclization cascade. This photocatalyst-free transformation avoids the cumbersome process of photocatalyst removal and potential toxicity. Mechanistic experiments revealed the generation of an electron donor-acceptor (EDA) complex during the reaction process. Moreover, the synthetic value of this protocol was further demonstrated through gram-scale reactions, conversion of the product, and total synthesis of an antidepressant drug.

Mechanistic studies reveal that the reaction is initiated by the addition of an arylsulfonyl radical to an alkyne from sulfur dioxide, followed by vinyl radical cyclization to afford the desired sulfonated dibenzazepines.
