Diagnostic yield of clinical exome sequencing in congenital hypogonadotropic hypogonadism considering the degree of olfactory impairment

Summary: This study aimed to evaluate the diagnostic yield of NGS in patients with CHH and explore the analysis method of selecting genes based on microsmia. The results showed that the virtual panel method confirmed the diagnosis of CHH in 5 patients, while selecting genes based on microsmia did not improve the diagnostic yield. Therefore, analyzing all genes related to CHH may be the most effective approach.

Summary: Kallmann syndrome (KS) is a rare genetic disorder characterized by idiopathic hypogonadotropic hypogonadism associated with anosmia. A novel noncanonical splice site variant in the ANOS1 gene was identified in two siblings with KS, leading to a premature termination of translation and a truncated anosmin-1 protein. This study highlights the importance of considering noncanonical splice site variants in molecular genetic diagnostics of KS.

Summary: In a cohort of eight families (11 patients) with autosomal recessive retinitis pigmentosa (arRP) associated with mutations in CNGB1, the study clinically characterized the disease and found new CNGB1 variants. The findings showed widespread retinal degeneration and variable penetrance and expressivity of the disease in these patients.

Summary: This study reported a rare case of co-occurrence of KS and WS2 due to SOX10 mutations. The patient presented with symptoms of olfactory loss, hypogonadism, deafness, and abnormal pigmentation. SOX10 mutation may be a common pathogenic factor for both KS and WS.

Kallmann syndrome in a patient with Weiss- Kruszka syndrome and a de novo deletion in 9q31.2

Summary: This report describes a patient with Weiss-Kruszka syndrome and Kallmann syndrome, who has a de novo 2.38 Mb heterozygous deletion in the chromosome 9q31.2 region. The deletion involves the loss of six protein-coding genes, with ZNF462 haploinsufficiency possibly contributing to the patient's phenotype but not explaining the Kallmann syndrome. Further analyses did not identify rare sequence variants in known Kallmann syndrome genes or aberrant splicing in expressed KS genes, suggesting a complex genetic mechanism underlying the patient's condition.
