Hybrid resolution molecular dynamics simulations of amyloid proteins interacting with membranes

Summary: A hybrid resolution molecular dynamics simulation method has been developed to investigate the interaction of amyloid proteins with lipid membranes, accelerating the aggregation propensity of amyloid proteins. This method allows for routine and accurate simulation of multi-protein and complex membrane systems, mimicking biologically relevant lipid membranes on microsecond time scales. It has been successfully applied to model the interactions of alpha-synuclein protein with a mixed lipid bilayer.

Misfolding and Self-Assembly Dynamics of Microtubule-Binding Repeats of the Alzheimer-Related Protein Tau

Summary: Pathological aggregation of intrinsically disordered tau protein, driven by the interactions between microtubule-binding (MTB) domains, is associated with Alzheimer's disease. Our results from atomistic discrete molecular dynamics simulations show that all four repeat monomers of tau are dynamic, with R3 displaying the strongest aggregation propensity. R1 is the least prone to aggregation, while R2 and R4 dimers contain both helix and beta-sheet structures.
