This study provides a comprehensive protocol to visualize and quantify lipid droplets in mouse muscle satellite cells, which can be adapted for other cell types of interest.
Lipid droplets are emerging as an important and dynamic organelle whose meta-bolism controls stem cell behavior. Here we present a comprehensive protocol to visualize and quantify these organelles in mouse muscle satellite cells (MuSCs). This protocol includes steps for BODIPY/LipidSpot610 staining of freshly iso-lated MuSCs, in vitro cultured myoblasts, and single myofibers to label lipid droplets and subsequent analysis and quantification of fluorescence signals. This protocol can be modified to stain lipid droplets in other cell types of interest. For complete details on the use and execution of this protocol, please refer to Yue et al. (2022).1
