Adjuvants and Vaccines Used in Allergen-Specific Immunotherapy Induce Neutrophil Extracellular Traps

Summary: Aluminum hydroxide and MPLA, as adjuvants in AIT vaccines, have varying effects on NET release by neutrophils, with aluminum being a more potent trigger and MPLA inducing stronger NET responses. The interaction of NETs and aluminum increased the expression of antigen-presenting molecules in monocytes and led to individual differences in IL-1β and IL-6 responses. These findings suggest that adjuvant-induced NETs may have autonomous and cooperative effects on early innate responses, potentially impacting the therapeutic efficacy of adjuvants and AIT vaccines.
