Genetic variation in endocannabinoid signaling is associated with differential network-level functional connectivity in youth

Lucinda M. Sisk et al.

Summary: Research has shown that individuals carrying the FAAH C385A polymorphism gene exhibit network-level changes in brain connectivity distinct from non-carriers, which may be related to genetic variations in endocannabinoid signaling, and these genotype-associated neural differences may emerge at a younger age.

Fatty acid amide hydrolase is lower in young cannabis users

Maya R. Jacobson et al.

Summary: Lower levels of FAAH enzyme were found in the brains of adult cannabis users during early abstinence, with higher cannabis use throughout the past year correlating with decreased [C-11]CURB binding. The study also detected lower [C-11]CURB binding in a younger cohort with less cumulative cannabis exposure, confirming and extending previous findings.

An adaptive randomized clinical trial in interstitial cystitis/bladder pain syndrome evaluating efficacy of ASP3652 and the relationship between disease characteristics and Hunner's lesions

Jos G. A. Houbiers et al.

Summary: This study evaluated the efficacy and safety of the fatty acid amide hydrolase (FAAH) inhibitor ASP3652 in patients with Interstitial Cystitis/Bladder Pain Syndrome (IC/BPS) and phenotyped patients based on Hunner's lesions (HL). ASP3652 was found to be safe and well tolerated in treatment, but did not demonstrate significant efficacy in IC/BPS. Differences observed between HL+ and HL- patients suggest that diagnosis and treatment of IC/BPS may need to be approached differently for these two phenotypes.

Summary: JNJ-42165279 appears to have an anxiolytic effect in subjects with social anxiety disorder (SAD), although trough concentrations with 25 mg once daily seemed insufficient to completely inhibit FAAH activity, resulting in suboptimal efficacy.

Summary: The study found that youth with the FAAH minor allele (AC or AA genotype) reported more drinking days, higher frequency of heavy episodic drinking, and more severe alcohol-related problems and consumption patterns. The association of FAAH C385A with drinking outcomes was mediated by coping motives.

CB2 Receptor in Microglia: The Guardian of Self-Control

Joanna Agnieszka Komorowska-Mueller et al.

Summary: Microglia are crucial for maintaining brain homeostasis, serving as both protectors against infections and potential hindrances to recovery when uncontrolled. Activating the endocannabinoid system, particularly CB2R, has been shown to reduce inflammation and hold promise for treating neuroinflammatory diseases. However, the precise mechanisms by which CB2R regulates microglial activity remain unclear, and ongoing research aims to further elucidate these pathways for potential therapeutic interventions.

Comorbid anxiety-like behavior in a rat model of colitis is mediated by an upregulation of corticolimbic fatty acid amide hydrolase

Haley A. Vecchiarelli et al.

Summary: Peripheral inflammatory conditions, such as those localized in the gastrointestinal tract, are highly comorbid with psychiatric disorders like anxiety and depression. These conditions are associated with sustained elevations in circulating glucocorticoid hormones, affecting central processes and regulating emotional behavior. The endocannabinoid system plays a significant role in modulating emotional behavior, and alterations in this system may contribute to the development of behavioral comorbidities in response to peripheral inflammation.

Fatty acid amide hydrolase binding is inversely correlated with amygdalar functional connectivity: a combined positron emission tomography and magnetic resonance imaging study in healthy individuals

Summary: This study revealed a negative correlation between levels of the endocannabinoid enzyme fatty acid amide hydrolase (FAAH) in the amygdala and functional connectivity in frontoamygdalar circuits involved in stress and affect control. The findings provide preliminary evidence supporting the role of FAAH in regulating brain circuits underlying fear and emotion processing in humans.

Fear extinction learning and anandamide: an fMRI study in healthy humans

Jennifer Spohrs et al.

Summary: Anxiety- and trauma-related disorders are severe illnesses with high prevalence, and the endocannabinoid system (ECS) has become a focus in psychopharmacological research. Studies show anxiolytic effects of endocannabinoids in rodent models, but research in humans remains scarce. Through research on healthy males, it was found that AC-heterozygotes of the SNP rs324420 have higher AEA levels, potentially related to fear extinction learning.
