Hierarchically Porous Biocatalytic MOF Microreactor as a Versatile Platform towards Enhanced Multienzyme and Cofactor-Dependent Biocatalysis

Summary: Hierarchically porous MOFs were constructed by controlled structural etching to enhance multienzyme and cofactor-dependent enzyme biocatalysis, which provided sufficient space for enzymes to reorientate and spread within MOFs and effectively accelerate the diffusion rate of reactants and intermediates, showing outstanding tolerance to inhospitable surroundings and recyclability.

Less Unfavorable Salt Bridges on the Enzyme Surface Result in More Organic Cosolvent Resistance

Summary: A smart salt bridge design strategy was used to improve the organic solvent resistance and thermostability of Bacillus subtilits Lipase A (BSLA), resulting in variants with significantly enhanced resistance towards organic solvents and improved thermal stability. Molecular dynamics simulations showed that locally refined flexibility and strengthened hydration played key roles in the increased resistance in organic solvents and at elevated temperatures. This salt bridge redesign approach could be a powerful tool for protein engineers to design lipases and other alpha/beta-hydrolases with enhanced resistance to organic solvents and/or thermal stability.

Fine-Tuning the Micro-Environment to Optimize the Catalytic Activity of Enzymes Immobilized in Multivariate Metal-Organic Frameworks

Summary: This study focuses on modifying the functionality and ratio of linkers in ZIFs using the MTV approach to optimize the activity of encapsulated enzymes. The research revealed a nonlinear correlation between enzyme activity and the ratio of hydrophilic linkers in MTV-ZIF-8, indicating the possibility of changing enzyme structure through conformation changes. The optimized BCL@ZIF-8 shows improved thermal and chemical stability compared to free BCL in solution, and also doubles the catalytic reactivity in kinetic resolution reactions.
