Dendritic Cells and Their Immunotherapeutic Potential for Treating Type 1 Diabetes

Farhan Ullah Khan et al.

Summary: Type 1 diabetes is caused by the destruction of pancreatic beta cells mediated by T cells. Dendritic cells play a crucial role in the initiation and development of this disease by presenting antigens to activate and regulate the immune response. Recent advancements in understanding dendritic cell function and regulation have led to the development of potential therapeutic strategies for treating type 1 diabetes.

Exosome loaded immunomodulatory biomaterials alleviate local immune response in immunocompetent diabetic mice post islet xenotransplantation

M. Rezaa Mohammadi et al.

Summary: In a study, hybrid alginate microcapsules (AlgXOs) releasing exosomes derived from human umbilical cord mesenchymal stem cells (XOs) were shown to attenuate immune response post-implantation. The XOs suppressed local immune microenvironment and immune cell activity, leading to significant efficacy in a mouse model of Type 1 Diabetes. The results suggest that the controlled release of XOs from AlgXOs could alleviate the local immune response to implantable biomaterials.

Summary: Insights into the etiology of type 1 diabetes (T1D) have shown a clear connection between genetic susceptibility and immune cell function, particularly regulatory T-cell (Treg) function. Advances in epigenetic and transcriptional analyses have provided evidence for Treg dysfunction in T1D, with future studies focusing on directly assessing Treg function and underlying mechanisms in T1D at tissue sites like pancreatic lymph nodes and pancreas.

Advances in Encapsulation and Delivery Strategies for Islet Transplantation

Siying Wu et al.

Summary: Type 1 diabetes mellitus is a chronic metabolic disease caused by autoimmune reactions leading to the destruction of pancreatic beta-cells. Islet transplantation has been recognized as a promising strategy for its curative treatment, but faces challenges such as donor shortage, poor revascularization, and inflammatory reactions. Biomaterial-based encapsulation and delivery strategies show promise in overcoming these obstacles and improving transplantation outcomes.

Immune Protection of Stem Cell-Derived Islet Cell Therapy for Treating Diabetes

Meghan Tahbaz et al.

Summary: Insulin injection and human pancreatic islet transplantation are current main therapies for T1D or late-stage severe T2D, but limitations such as islet shortage and immunosuppression side effects call for the need to develop novel methods for generating protected functional human islets. Human pluripotent stem cells offer a potentially limitless cell source, and studying immune tolerance induction in hPSC-derived human pancreatic islets contributes to the goal of curing insulin-dependent diabetes.

Summary: Islet transplantation is a promising strategy for treating type 1 diabetes by replacing damaged beta-cells with new islets in a minimally invasive procedure. However, it faces significant drawbacks such as long-term immunosuppression and a lack of compatible donors. Biomaterial-assisted islet transplantation, particularly using hydrogels, is an emerging technology that aims to overcome these challenges and improve beta-cell viability, decrease inflammatory responses, and enhance insulin secretion.
