Photocaged 5′ cap analogues for optical control of mRNA translation in cells

Nils Kloecker et al.

Summary: Translation of mRNA is a crucial process in gene expression, and a new study introduces FlashCaps, cap analogues with photo-cleavable groups, that can regulate translation of mRNA effectively. These compounds inhibit translation in vitro and in mammalian cells, but can be restored through irradiation for efficient translation. FlashCaps present a promising approach for controlling mRNA expression and dosing mRNA therapeutics with spatial and temporal control.

Enzymatic Generation of Double-Modified AdoMet Analogues and Their Application in Cascade Reactions with Different Methyltransferases

Mehmet Erguven et al.

Summary: Methyltransferases and methionine adenosyltransferases play important roles in biocatalytic reactions, allowing for the transfer of functional moieties. However, current methods limit selective modification in mixtures. The use of AdoMet analogues with additional modifications can improve targeting selectivity of methyltransferases.

A second type of N7-guanine RNA cap methyltransferase in an unusual locus of a large RNA virus genome

Ashleigh Shannon et al.

Summary: In this study, we discovered that the ORF1a region of certain members of the large genome Tobaniviridae family indeed harbors N7-specific methyltransferase activity. We presented its structure as the first N7-specific Rossmann-fold (RF) methyltransferase identified for (+)RNA viruses, which is remarkably different from that of the known Coronaviridae ORF1b N7-MTase gene. The appearance of this methyltransferase in an unexpected location in the genome introduces a split-off in the classification of Tobaniviridae.

2′-O-Methylation of the second transcribed nucleotide within the mRNA 5′ cap impacts the protein production level in a cell-specific manner and contributes to RNA immune evasion

Karolina Drazkowska et al.

Summary: This study explores the impact of nucleotide modifications in mRNA capping on protein production levels and immune response. The researchers find that 2'-O-methylation of the second transcribed nucleotide can significantly affect protein synthesis in a cell-specific manner and is linked to transcript escape from the host immune response.

Engineered SAM Synthetases for Enzymatic Generation of AdoMet Analogs with Photocaging Groups and Reversible DNA Modification in Cascade Reactions

Freideriki Michailidou et al.

Summary: Exploring the methylation and demethylation of DNA, RNA, and proteins as a major regulatory mechanism benefits from the generation of AdoMet analogs with photocaging groups, allowing for site-specific inhibition and timed removal of methyltransferases. The engineering of SAM synthetase from Cryptosporidium hominis enables the efficient generation of non-aliphatic AdoMet analogs, which can be used for sequence-specific modification and light-triggered removal of DNA and RNA modifications.
