A miRNA-based gene therapy nanodrug synergistically enhances pro-inflammatory antitumor immunity against melanoma

Yawen Ma et al.

Summary: MicroRNA-based gene therapy using biocompatible nanocarriers shows promise in treating ocular melanoma by up-regulating tumor suppressor miR-30a-5p and inhibiting malignant phenotypes. The nanocarrier with high miRNA loading and redox-responsive release capabilities enhances the therapeutic effect. Furthermore, it promotes M1 macrophage polarization and activates Fenton reaction, synergistically enhancing anti-tumor immunity. The study demonstrates the potential of this nanocarrier for targeted gene therapy in melanoma and other cancers.

Inducible MicroRNA-132 Inhibits the Production of Inflammatory Cytokines by Targeting TRAK TAK1, and TAB1 in Teleost Fish

Junxia Cui et al.

Summary: The innate immune response is crucial for defending against pathogen infection, but excessive inflammation can lead to inflammatory and autoimmune diseases. Recent evidence suggests that miRNAs can act as negative regulators in the regulation of inflammation and immune responses in mammals. However, little is known about miRNA-mediated immune regulation networks in lower vertebrates. This study identifies a miRNA, miR-132, in miiuy croaker that acts as a negative regulator in the host's bacterium-induced inflammatory response. MiR-132 inhibits the production of inflammatory cytokines by targeting multiple factors, thus preventing excessive inflammation.

miRNAs in the regulation of mTOR signaling and host immune responses: The case of Leishmania infections

Sajad Rashidi et al.

Summary: Micro RNAs (miRNAs) play a role in regulating gene expression and can interact with cell signaling, influencing the pathogenesis of different diseases. The crosstalk between miRNAs and cellular signaling networks, including mTOR, can impact disease progression. While there have been studies on the crosstalk between miRNAs and mTOR signaling in various diseases, research on parasitic infections is limited. Targeting miRNAs as regulators of mTOR could provide new therapeutic strategies against parasitic diseases.

Serum-derived exosomes induce proinflammatory cytokines production in Cynoglossus semilaevis via miR-133-3p

Zhanpeng Sun et al.

Summary: Exosomes are small membrane-enclosed vesicles secreted by various types of cells. In this study, serum-derived exosomes from Chinese tongue sole were isolated and examined for their effects on inflammatory processes following Vibrio harveyi infection. The results showed that exosomes increased in abundance with bacterial infection time, leading to changes in inflammatory cytokine expression. miRNA profiling revealed that miR-133-3p was significantly more abundant in exosomes from infected fish, and it was found to inhibit NF-kappa B signaling pathways to promote inflammatory responses.
