Kinetics of elementary steps in loop-mediated isothermal amplification (LAMP) show that strand invasion during initiation is rate-limiting

Summary: Loop-mediated isothermal amplification (LAMP) is easier to implement than PCR for point-of-care diagnostic tests. This study characterizes the kinetics of the elementary steps in LAMP and shows that the rate-limiting step is strand invasion / initiation. The loop primer accelerates the rate of initiation and Bst-LF polymerase demonstrates fast and processive strand displacement synthesis. These findings contribute to a kinetic model that can be used to engineer more sensitive and faster LAMP reactions, with potential applications for other isothermal DNA amplification methods.

Summary: A loop-mediated isothermal amplification (LAMP) assay was developed for the rapid detection of Colletotrichum gloeosporioides, the causal agent of anthracnose in fruits and vegetables. The assay targeted the upregulated transcript enoyl-CoA hydratase (ECH) during the quiescent stage of C. gloeosporioides. The LAMP assay provided a naked-eye detection within 23 minutes and exhibited high specificity.

Establishment of a rapid method for skipjack tuna (Katsuwonus pelamis) authentication using molecular beacons in loop-mediated isothermal amplification

Summary: One major drawback to the traditional loop-mediated isothermal amplification (LAMP) detection methods is the increased likelihood of detecting false-positive signals derived from non-specific amplification. Molecular beacon (MB) is increasingly being used in many applications and the MB-LAMP assay has proved itself as a target specific method. The present work selected skipjack tuna as a case study, and developed a novel MB-LAMP assay for rapid species authentication. The assay demonstrated high specificity and could detect as little as 0.5 pg of skipjack tuna DNA.

Development of duplex real-time polymerase chain reaction for simultaneous detection of oilfish- and escolar-derived components

The method showed high specificity without cross-reaction on other species, with a limit of detection for DNA of 0.0002 ng μL(-1) for R. pretiosus and 0.002 ng μL(-1) for L. flavobrunneum. Oilfish- and escolar-derived components were detected in 12 of the 70 commercial samples, consistent with DNA barcoding test results.
