Simultaneous Dual-Sensing Platform Based on Aptamer-Functionalized DNA Hydrogels for Visual and Fluorescence Detection of Chloramphenicol and Aflatoxin M1

Summary: A chloramphenicol and aflatoxin M1 aptamer-functionalized DNA hydrogel was designed for the simultaneous detection of chloramphenicol and aflatoxin M1. By using acrydite-modified chloramphenicol aptamer sequence, the DNA hydrogel was synthesized and used for visual detection of chloramphenicol. The DNA hydrogel was formulated with 60% linear polyacrylamide-DNA conjugate, 40% acrylamide and chloramphenicol aptamer/DNA strand-1 at a molar ratio of 1:1, with the lowest concentration of chloramphenicol for gel dissociation being 1.0 nM at 25 degrees C. The formalized aptamer-functionalized DNA hydrogel was also used to detect aflatoxin M1 by measuring the recovery of fluorescence signal. The detection platform showed promising results for the simultaneous detection of chloramphenicol and aflatoxin M1 in aqueous solution and milk.

Aflatoxin B1 exposure induced developmental toxicity and inhibited muscle development in zebrafish embryos and larvae

Summary: The prevalence of Aflatoxin B1 (AFB1), a highly toxic mycotoxin, is increasing worldwide and poses various health problems. The direct toxicity of AFB1 on embryonic development, especially muscle development, has not been extensively studied. This study used zebrafish embryos as a model to investigate the mechanism of AFB1's direct toxicity on embryonic development, revealing inhibition of muscle development through oxidative damage, apoptosis, and disruption of tight junction complexes (TJs).

Total Flavonoids of Rhizoma Drynariae Mitigates Aflatoxin B1-Induced Liver Toxicity in Chickens via Microbiota-Gut-Liver Axis Interaction Mechanisms

Summary: This study investigated the protective effects and potential mechanisms of total flavonoids of Rhizoma Drynaria (TFRD), a traditional Chinese medicinal herb, against Aflatoxin B1 (AFB1)-induced liver injury. The results showed that TFRD supplementation improved intestinal permeability, corrected changes in gut microbiota and liver damage, and improved plasma metabolites affected by AFB1 exposure. Furthermore, TFRD treatment suppressed oxidative stress, hepatic lipid deposition, and hepatic ferroptosis gene expression. These findings suggest that TFRD can potentially counteract the detrimental effects of mycotoxins.

AFB1 colorimetric aptamer sensor for the detection of AFB1 in ten different kinds of miscellaneous beans based on gold nanoparticles and smartphone imaging

Summary: A simple and low-cost smartphone-assisted assay was developed for measuring aflatoxin B1 in miscellaneous beans. The assay utilized a modified QuEChERS method with aptamers and gold nanoparticles as recognition elements and indicators, respectively. The assay showed good sensitivity and specificity, with a linear range of 0.2-8.0 ng/g and a detection limit of 0.08 ng/g. The recovery rate of aflatoxin B1 in bean samples ranged from 87.18% to 110.24%. The entire process took only 15 minutes. This colorimetric aptamer sensor can be used for quick aflatoxin B1 detection in beans.

Rapid and label-free detection of aflatoxin B1 using a rationally truncated aptamer and via circular dichroism measurement

Summary: We present a simple aptamer assay for rapid analysis of the carcinogenic fungal metabolite AFB1. By rationally truncating the original anti-AFB1 aptamer, a short aptamer with improved performance was obtained. Different concentrations of AFB1 were detected by measuring the circular dichroism spectra of this short aptamer. The detection limit reached 0.6 nM, which was lower than that of the original aptamer (80 nM).
