XBP1 Activation Reduces Severity of Polycystic Kidney Disease due to a Nontruncating Polycystin-1 Mutation in Mice

Matteus Krappitz et al.

Summary: The study found that the expression of active XBP1 in a murine model with missense Pkd1 mutations can increase the levels of mutant PC1 and improve its trafficking in cells. Transgenic expression of spliced XBP1 can reduce cell proliferation and improve ADPKD progression in tubule segments destined to form cysts.

More dissimilarities than affinities between DNAJB11-PKD and ADPKD

Isabella Pisani et al.

Summary: DNAJB11-PKD is an atypical form of polycystic kidney disease that shows distinct clinical features compared to the traditional ADPKD. Patients with DNAJB11-PKD have smaller kidneys, smaller cysts, and a slower progression to end-stage kidney disease. They also display proteinuria, higher prevalence of type 2 diabetes mellitus and nephrolithiasis, but lower prevalence of cardiac valvular defects.

The molecular mechanism and functional diversity of UPR signaling sensor IRE1

Samirul Bashir et al.

Summary: The endoplasmic reticulum is responsible for protein folding and maturation, but can be affected by various stress conditions triggering the Unfolded Protein Response. This response involves the activation of sensors like IRE1 which signal to restore cellular homeostasis, and IRE1 can switch its substrate specificity to cotransnationally transported mRNAs in a process known as Regulated IRE1 Dependent Decay (RIDD).

Bi-allelic pathogenic variations in DNAJB11 cause Ivemark II syndrome, a renal-hepatic-pancreatic dysplasia

Penelope Jordan et al.

Summary: Research has shown that biallelic pathogenic variations in DNAJB11 can lead to a severe fetal disease characterized by enlarged cystic kidneys, dilation and proliferation of pancreatic duct cells, and liver ductal plate malformation. The disease is associated with Ivemark II syndrome and kidney cysts developed exclusively from a specific tubular segment. This recessive disease is categorized as a ciliopathy rather than a disease within the autosomal dominant tubulointerstitial kidney disease spectrum.
