Inactivation of Pseudomonas aeruginosa biofilms by thymoquinone in combination with nisin

Hong Chen et al.

Summary: This study evaluated the synergistic effect of thymoquinone and nisin in reducing biofilm formation of P. aeruginosa on lettuce, and investigated their anti-virulence and anti-biofilm mechanisms. Thymoquinone inhibited the production of autoinducers and virulence factors, and enhanced the susceptibility of P. aeruginosa biofilms to nisin. It disrupted the quorum sensing system, altered cell membrane component, and down-regulated the expressions of genes related to virulence, efflux pump, and antioxidation.

ROS-induced lipid peroxidation modulates cell death outcome: mechanisms behind apoptosis, autophagy, and ferroptosis

Bingqing Wang et al.

Summary: Reactive oxygen species (ROS) play a crucial role in cell death processes such as apoptosis, autophagy, and ferroptosis through lipid peroxidation. The oxidation of phospholipid bilayers can trigger mitochondrial apoptosis and endoplasmic reticulum stress, while also affecting autophagy. Additionally, ROS and lipid peroxides are involved in ferroptosis, and the nuclear factor erythroid 2-related factor 2 regulates ferroptosis under oxidative conditions. This review provides insights into the interplay among apoptosis, autophagy, and ferroptosis mediated by ROS and lipid peroxidation, offering new directions for studying diseases related to pathological cell death.

Synthesis, antimicrobial, antibiofilm and computational studies of isatin-semicarbazone tethered 1,2,3-triazoles

Aman Kumar et al.

Summary: In this study, a new class of isatin-semicarbazone tethered 1,2,3-triazole hybrids was synthesized via Cu(I)-mediated azide alkyne cycloaddition reaction. The structural characteristics of the compounds were identified using various spectral techniques, and the synthesized derivatives were tested for antimicrobial and antibiofilm activity. The results showed that the triazole hybrids exhibited promising antimicrobial and antibiofilm properties, making them potential candidates for biomaterial applications.

Blockage of autophagosome-lysosome fusion through SNAP29 O-GlcNAcylation promotes apoptosis via ROS production

Francesca Romana Pellegrini et al.

Summary: This study identified a new pathway in autophagy connecting O-GlcNAcylated SNAP29 to autophagic flux blockage and autophagosome accumulation, which leads to ROS production and apoptotic cell death. These findings provide a new opportunity for therapeutic intervention in cancer and other autophagy-related diseases.

Tetrahedral framework nucleic acids alleviate irradiation-induced salivary gland damage

Xueping Xie et al.

Summary: In this study, the role of tFNAs in irradiation-induced salivary gland damage was investigated. The results showed that tFNAs could promote cell proliferation, inhibit cell apoptosis, and reduce cell death caused by irradiation. Mechanism studies revealed that tFNAs inhibited cell apoptosis by regulating the Bcl-2/Bax/Caspase-3 signaling pathway and reducing inflammation-induced cell damage. Animal experiments showed that tFNAs could alleviate weight loss, increased water intake, decreased saliva production, and prolonged salivation lag time caused by irradiation, and improve salivary gland damage.
