Combining LC-MS/MS profiles with network pharmacology to predict molecular mechanisms of the hyperlipidemic activity of Lagenaria siceraria stand

Summary: Lagenaria siceraria Stand. (bottle gourd) is traditionally used in Ayurvedic medicine for hypertension and obesity. This study investigated the lead compounds from bottle gourd and their mechanisms of action in obesity and hypertension. Metabolite screening, animal studies, and network pharmacology analyses were performed. The results showed that the compounds from bottle gourd improved lipid profiles in hyperlipidemic animals and acted through the insulin and ErbB signaling pathways. These findings suggest that bottle gourd could be a natural alternative for managing hyperlipidemia.

Lipid-induced monokine cyclophilin-A promotes adipose tissue dysfunction implementing insulin resistance and type 2 diabetes in zebrafish and mice models of obesity

Summary: Research suggests that the interaction between macrophages and adipocytes in obese individuals can lead to insulin resistance. This study found that a lipid-induced compound called CyPA significantly impairs adipocyte functions and insulin sensitivity. Inhibition of CyPA reduces adipose tissue inflammation and restores adipocyte function, improving insulin sensitivity. Mechanistically, the interaction between CyPA and its receptor CD147 activates a signaling pathway that promotes adipocyte inflammation and inhibits adipocyte differentiation. Inhibiting CyPA or CD147 can restore adipose tissue function and insulin sensitivity.

Summary: This study assessed the effects of pigmented and non-pigmented rice, as well as an extruded food product, on DPP-4 inhibition and GLP-1 secretion. The results showed that the ethanol/water extract of rice bran had the highest DPP-4 inhibitory activity, while the extruded products stimulated GLP-1 secretion mostly through n-hexane or ethanol extraction. The extruded food items also had a lower arsenic content.

The PD leaf extracts can improve glucose metabolism and alleviate oxidative stress in rats.
