Epitopes screening and vaccine molecular design of SADS-CoV based on immunoinformatics

Shinian Li et al.

Summary: This study aims to predict the epitope of S protein B cells and T cells (MHC class I and class II) through immunoinformatics methods, screening and identifying protective antigenic epitopes that induce major neutralized antibodies and activate immune responses to construct epitope vaccines. The results show that the constructed epitope vaccine interacts stably with TLR-3 receptors and has good immunogenicity, effectively activating humoral and cellular immunity. Therefore, the constructed epitope vaccine will provide a new theoretical basis for the design of SADS-CoV antiviral drugs and related research on coronaviruses such as SARS-CoV-2.

Development of mRNA Vaccines/Therapeutics and Their Delivery System

Sora Son et al.

Summary: The rapid development of mRNA vaccines has been useful in managing COVID-19 and may be beneficial in managing future infectious disease outbreaks. mRNA vaccines can easily alter target antigens by changing the coding region sequence, making them suitable for rapidly evolving outbreaks. They have the potential to induce successful antigen-specific immunity by expressing target antigens in cells and triggering immune responses. The approved COVID-19 mRNA vaccines have shown significant efficacy in preventing infections. mRNA's ability to produce defective target proteins has opened possibilities for treating intractable diseases. Clinical applications of mRNA vaccines/therapeutics require safe delivery strategies. This review summarizes current knowledge on mRNA vaccines/therapeutics, their clinical applications, and delivery strategies.

Summary: The study shows that species-specific CMV variant-specific antibody responses contribute to the immunogenicity of HCMV vaccines, indicating the necessity of considering pre-existing immunity in non-human primate models and the impact of different species' immune responses post-vaccination.

Safety and immunogenicity of an mRNA-lipid nanoparticle vaccine candidate against SARS-CoV-2 A phase 1 randomized clinical trial

Peter G. Kremsner et al.

Summary: The RNActive technology platform was used to prepare CVnCoV, a COVID-19 vaccine containing optimized mRNA encoding for a stabilized form of SARS-CoV-2 spike protein. The dosage escalation phase 1 study showed that 12 µg of CVnCoV vaccine elicited immune responses comparable to those seen in convalescent sera from COVID-19 patients, with acceptable safety and reactogenicity.

mRNA therapy restores euglycemia and prevents liver tumors in murine model of glycogen storage disease

Jingsong Cao et al.

Summary: Glycogen Storage Disease 1a (GSD1a) is a rare inherited metabolic disorder characterized by hypoglycemia and high risk of liver cancer. In this study, researchers developed an mRNA-based G6Pase-alpha delivery therapy that was shown to be efficacious and safe in a mouse model of GSD1a.

Leukocyte trafficking to the lungs and beyond: lessons from influenza for COVID-19

Ronen Alon et al.

Summary: The understanding of COVID-19 clinical manifestations requires knowledge of how immune cells are recruited to virus-infected lungs and how this recruitment varies among individuals. The orchestration of leukocyte trafficking to respiratory system is controlled by specific molecules, which can lead to pathological complications when uncontrolled. The incomplete understanding of trafficking signals in response to SARS-CoV-2 infection calls for further research for potential therapeutic interventions.
