Evaluation of the effects of natural isoquinoline alkaloids on low density lipoprotein receptor (LDLR) and proprotein convertase subtilisin/kexin type 9 (PCSK9) in hepatocytes, as new potential hypocholesterolemic agents

Binita Maharjan et al.

Summary: Nine different isoquinoline alkaloids were evaluated for their effects on a cellular model of hepatocyte. Berberine, californidine, and govaniadine showed similar effects as simvastatin on low density lipoprotein receptor (LDLR) expression. However, californidine and berberine reduced the expression of proprotein convertase subtilisin/kexin type 9 (PCSK9), while govaniadine increased both LDLR and PCSK9 levels. Furthermore, all compounds were able to reduce total cholesterol levels in hepatocytes.

Fengping Chen et al.

Summary: Ascorbic acid (VC) and transferrin (TRF) form a complex through hydrogen bonds and van der Waals force, altering the secondary structure of TRF. VC binding to TRF enhances its scavenging capacity on superoxide anion radicals in low concentrations, but inhibits its ability against DPPH radicals after complex formation.

Summary: A Rhodamine-based fluorescent probe R1 has been successfully used to detect BSA and HHb proteins through non-covalent bonding. The interaction between R1 and proteins has been revealed by changes in absorption, emission, and circular dichroism spectra. Experimental results have shown a moderate binding strength between proteins and R1 through static quenching mechanism. The hyperchromism in the absorption band of proteins with R1 has provided vital information, and the binding affinity values have been calculated from Stern-Volmer plots.
