One-step competitive assay for detection of thrombin via disassembly of diblock oligonucleotide functionalised nanogold aggregates

Summary: In this study, a simple, sensitive, and cost-effective sensing strategy for thrombin was developed. The strategy involved the use of polyadenine diblock oligonucleotides anchored on gold nanoparticles (AuNPs) to detect thrombin. Binding of thrombin to its aptamer triggered the disassembly of AuNP aggregates, resulting in a significant signal change. The assay demonstrated a detection limit of 0.81 pM and a dynamic range from 10 pM to 10 nM, and could be performed in diluted serum solutions without sophisticated instruments.

In-situ synthesis of carbon dots embedded wrinkled-mesoporous silica microspheres for efficiently capturing and monitoring organochlorine pesticides from water and fruit juice

Summary: This study proposed an in-situ synthesis strategy to fabricate carbon dots embedded wrinkled mesoporous silica (CDs/WMS) microspheres. The CDs doping allowed the microspheres to maintain their wrinkled and highly mesoporous structures. The CDs/WMS composite was used as a solid phase microextraction (SPME) coating for sensitive determination of ultra-trace organochlorine pesticides (OCPs) from aqueous solutions. Under optimized conditions, the proposed analytical method showed wide linearity range, low detection limits, and satisfactory reproducibility, and was successfully applied to real water and fruit juice samples.

Synthesize of the chitosan-TPP coated betanin-quaternary ammonium-functionalized mesoporous silica nanoparticles and mechanism for inhibition of advanced glycation end products formation

Summary: In this study, betanin was encapsulated in chitosan-sodium tripolyphosphate coated quaternary ammonium-functionalized mesoporous silica nanoparticles (CS@QAMSNPs) to enhance its ability to inhibit the formation of advanced glycation end products (AGEs). The betanin-CS@QAMSNPs showed a higher inhibition rate of AGEs formation (70.29%) compared to betanin alone (39.48%). They also exhibited better trapping of methylglyoxal and retained antioxidant capacity under high temperatures. Furthermore, the betanin-CS@QAMSNPs reduced the average degree of substitution per peptide molecule value (DSP) of glycation sites in bovine serum albumin and demonstrated good biocompatibility.

Facile immunochromatographic assay based on metal-organic framework-decorated polydopamine for the determination of hydrochlorothiazide adulteration in functional foods

Summary: A novel immunochromatographic assay (ICA) based on metal-organic framework-decorated polydopamine (MOF@PDA) was developed for the determination of hydrochlorothiazide (HCTZ) adulteration in functional foods. The MOF@PDA carrier showed a high coupling rate of 91.7% with HCTZ antibody. The developed MOF@PDA-ICA exhibited sensitivity 27-fold higher than the reported ICA, with detection limits of 5.93 μg/kg in functional tablets and 4.72 μg/kg in capsules. The parallel tests with commercial functional tablets and capsules demonstrated the consistent results between MOF@PDA-ICA and LC-MS/MS (R2 > 0.99).

Dual-template molecularly surface imprinted polymer on fluorescent metal-organic frameworks functionalized with carbon dots for ascorbic acid and uric acid detection

Summary: In this research, a dual-template molecularly imprinted polymer surface was prepared on a blue fluorescent Cr-based MOF functionalized with yellow emissive carbon dots (Y-CDs), using L-ascorbic acid (AA) and uric acid (UA) as templates for simultaneous selective recognition of AA and UA. The developed nanocomposite probe (Y-CDs/Cr-MOF@MIP) contains two recognition site cavities and emits dual fluorescence spectra - blue emission from Cr-MOF and yellow emission from Y-CDs. The quenching of Y-CDs' yellow fluorescence and Cr-MOF's blue fluorescence in the presence of AA and UA, respectively, allows for the design of a dual-template biosensor for simultaneous detection of AA and UA. The sensor displayed a dynamic linear response and high selectivity and sensitivity for AA and UA detection in human urine samples.

Proximity binding-initiated DNA walker and CRISPR/Cas12a reaction for dual signal amplification detection of thrombin

Summary: We have developed a highly sensitive fluorescent thrombin biomarker sensing method by combining the DNA walker and CRISPR/Cas12a system. This method utilizes the DNA moving arms to localize on AuNP tracks and generate CRISPR/Cas12a-responsive dsDNAs, which restores the fluorescence by pushing the dye away from AuNPs. The method has a detection limit of 29.5 fM and has been verified for selectively detecting thrombin against interference proteins and in diluted serums, making it a promising approach for disease diagnosis.

Highly sensitive protein detection by aptamer-based single-molecule kinetic fingerprinting

Nucleic acid aptamers generated by SELEX have shown promise as an alternative to immunoreagents for detection, offering advantages in synthesis and reproducibility. SiMREPS, a single molecule recognition platform, allows for high sensitivity detection of biomarkers without requiring high-affinity probes. This study demonstrates the applicability of aptamers as detection probes in SiMREPS for two clinically relevant biomarkers, VEGF165 and IL-8, with detection limits in the low femtomolar range.
