Cholesterol modulates the physiological response to nanoparticles by changing the composition of protein corona

Summary: The metabolome profile is an underexplored factor that affects the efficacy and safety of nanomedicines; using cholesterol as an example, this study demonstrates the importance of developing personalized nanomedicines by targeting disease-related metabolites. Cholesterol levels can impact the generation of the protein corona, leading to enhanced inflammatory responses in macrophages and increased cellular uptake in hepatocytes. In vivo biodistribution assays show that hypercholesterolemic mice have altered tissue delivery patterns of nanoparticles. This research highlights the need to consider the metabolome profile in developing nanomedicines for improved targeting efficacy and safety.

Monitoring the distribution of internalized silica nanoparticles inside cells via direct stochastic optical reconstruction microscopy

Summary: Understanding the precise localization of nanoparticles within cells is crucial for designing effective nanomedicines. This study utilized direct stochastic optical reconstruction microscopy (dSTORM) to determine the exact position of nanoparticles within cells, benefiting from its nanometric resolution, multicolour ability, and minimal invasiveness. The results showed that dSTORM provided higher spatial resolution compared to conventional fluorescence microscopy for locating nanoparticles within cells. Additionally, the distribution of nanoparticles in different cell lines over time was investigated.

The brush conformation of PEG enables stealth behavior of nanocarriers and inhibits uptake by phagocytic cells, facilitating long blood circulation and targeted delivery. The study demonstrates that, in addition to surface chemistry, the conformation of polymers plays a crucial role in controlling cellular interactions of nanocarriers.

Molybdenum derived from nanomaterials incorporates into molybdenum enzymes and affects their activities in vivo

Summary: Understanding the in vivo biotransformation of nanomaterials, such as molybdenum derived from MoS2, can shed light on their long-term effects on liver metabolism. Through a combination of in vivo experiments and molecular dynamics simulations, the study reveals that the biodistribution of molybdenum is mediated by protein coronas, mainly with apolipoprotein E, leading to increased enzyme activities in the liver. The findings suggest that nanomaterials consisting of essential trace elements may be converted into active biological molecules in organisms.
