Modified Greener Procedure for the Synthesis of Cucurbit[6]uril

Oleksandr Zbruyev et al.

Summary: A modified procedure for the synthesis of cucurbit[6]uril on the kilogram scale was developed, which involved reducing the amount of chemicals used in the reaction and simplifying the subsequent workup steps. This procedure not only improved the yield of the product but also made the synthesis more environmentally friendly.

Preparation, characterization, and biological activity study of thymoquinone-cucurbit[7]uril inclusion complex

Lubna Alrawashdeh et al.

Summary: The study investigated the host-guest inclusion complex between cucurbit[7]uril and thymoquinone in aqueous solution. Experimental techniques such as H-1 NMR and UV-visible spectroscopy confirmed the formation of a stable inclusion complex. The complexation stoichiometry was determined to be 1:1 with a moderate binding affinity, supported by both experimental and theoretical calculations. Furthermore, the cytotoxic effect of the CB[7]-TQ complex was enhanced against cancer cells, demonstrating the potential of CB[7] as a drug carrier with low toxicity.

Investigation of the Complexation between 4-Aminoazobenzene and Cucurbit[7]uril through a Combined Spectroscopic, Nuclear Magnetic Resonance, and Molecular Simulation Studies

Sabine Sarraute et al.

Summary: In this study, the inclusion complex formation between cucurbit[7]uril and 4-aminoazobenzene in aqueous solution was investigated using experimental techniques and molecular simulations. The results showed that the binding affinity was high in acidic solutions but no association was detected in neutral solutions. The thermodynamic properties of complex formation were studied and it was found that the high binding constant was attributed to the changes in ΔH-r(o) and ΔS-r(o) values. Molecular dynamics simulations were used to interpret the experimental results, taking into account the cis-trans isomerization of 4-aminoazobenzene.

SAMPL7 Host-Guest Challenge Overview: assessing the reliability of polarizable and non-polarizable methods for binding free energy calculations

Martin Amezcua et al.

Summary: The SAMPL challenges focus on testing and advancing computational methods for pharmaceutical drug discovery, particularly in predicting binding affinities. Challenges in predicting affinities include conformational sampling, protonation state uncertainties, variation in test sets, and lack of high quality experimental data. Results suggest that further exploration of polarizable force fields may be warranted in these challenges.

A reference scale of cucurbit[7]uril binding affinities

Mohammad A. Alnajjar et al.

Summary: Accurately determining ultra-high binding affinities in supramolecular host-guest chemistry is challenging due to sensitivity constraints of common titration methods. Competitive titrations with well-established reference compounds can provide a reference scale for determining high binding affinities, such as with the host cucurbit[7]uril (CB7) in the range from 10^3 to 10^15 M^-1. These commercially available reference compounds will aid in future research on stimuli-responsive host-guest systems.
