Molecular Dynamics and Multi-Spectroscopic of the Interaction Behavior between Bladder Cancer Cells and Calf Thymus DNA with Rebeccamycin: Apoptosis through the Down Regulation of PI3K/AKT Signaling Pathway

Summary: The interaction between Rebeccamycin and calf thymus DNA (ctDNA) in the absence and presence of H1 was investigated. The results showed that Rebeccamycin interacted with ctDNA through intercalator or binding modes in the absence of H1, while the presence of H1 resulted in revealing the intercalator and groove binding modes of the ctDNA-Rebeccamycin complex. The binding constants indicated strong binding of Rebeccamycin with ctDNA. The thermodynamic parameters suggested that both van der Waals forces and hydrogen bonds played predominant roles in the binding of Rebeccamycin to ctDNA in both situations. Circular dichroism results showed no major conformational changes in ctDNA upon interacting with Rebeccamycin. The relative viscosity of ctDNA increased, confirming the intercalation and groove binding modes of Rebeccamycin in the absence and presence of H1. Molecular docking simulation results corroborated the experimental findings and demonstrated the effectiveness of Rebeccamycin in inhibiting and proliferating T24 and 5637 cells. The ability of Rebeccamycin to inhibit cell proliferation and tumor growth through the induction of apoptosis by down-regulating the PI3K/AKT signaling pathway was also observed.

Molecular mechanism of high-pressure processing regulates the aggregation of major royal jelly proteins

Summary: This study evaluated the effect of high-pressure processing on the aggregation behavior of major royal jelly proteins (MRJPs) and revealed the regulation mechanism at the molecular level. The results showed that high-pressure processing effectively disrupted the filament aggregation of MRJPs and regulated their aggregation behavior mainly by affecting the higher-order structure. This study provides valuable insights and theoretical support into the regulation mechanism of food-derived protein aggregation and supports the pre-treatment of MRJPs in the development of functional foods.
