Design of a biocatalytic cascade for the enzymatic sulfation of unsulfated chondroitin with in situ generation of PAPS

Summary: Sulfation of molecules is crucial for the functionality of living organisms, especially in mammals where the sulfation of polysaccharides plays a key role in the formation of proteoglycans. These sulfated polysaccharides act as signal modulators in communication processes and are involved in various biological events. In order to develop sustainable sulfation processes, a multienzyme system was created to synthesize and couple the expensive and difficult-to-obtain PAPS to a chondroitin sulfation system. The resulting product was analyzed using NMR.

Structural analysis and mutagenesis suggested the importance of specific residues for the activity of ChSase B6. The characterization and structure of ChSase B6 provided guidance for its industrial application in chondroitin sulfate bioresource development.

In this study, we combined AlphaFold with crystal structures to investigate the structure of a previously unreported enzyme. We identified a functionally important structural element that was missing in the AlphaFold model but present in the crystal structure. We were able to reconstruct the protein oligomer using computational approaches and successfully solve the protein structure by phasing the diffraction data with the AlphaFold model. This work demonstrates the integration of AlphaFold with experimental data and its significance in protein analysis, crystallographic phasing, and structure solution.

While total synthesis and controlled depolymerization of natural fCS polysaccharides are the main approaches, limitations exist which could be overcome through a semisynthesis strategy involving regioselective fucosylation and sulfation of a microbial sourced polysaccharide lacking fucose and sulfate decorations. By preparing structurally homogeneous fCS isomers through a multistep process and conducting detailed structural characterization and conformational behavior studies, researchers have made significant progress in this field.
