Systems biology strategy and experimental validation to uncover the pharmacological mechanism of Xihuang Pill in treating non-small cell lung cancer

Zhenjie Zhuang et al.

Summary: The present study aimed to investigate the pharmacological mechanisms involved in treating non-small cell lung cancer (NSCLC) with Xihuang Pill (XHP), a representative anticancer Chinese patented medicine. XHP was found to suppress cancer cell growth in NSCLC by regulating pathways associated with cell proliferation and immunity. In addition, XHP may increase the expression of CACNA1C to suppress immune cell infiltration and regulate the expression of checkpoint-related genes, thereby improving the overall survival of NSCLC patients.

Xihuang pill in the treatment of cancer: TCM theories, pharmacological activities, chemical compounds and clinical applications

Hong-Bin Xu et al.

Summary: This study aims to summarize recent advances in research on the anti-cancer efficacies of Xihuang pill, a famous traditional Chinese formula, based on the theoretical basis of traditional Chinese medicine, pharmacological activities, chemical components, and clinical application. The study found that Xihuang pill exerts anti-cancer effects by inducing cancer cell apoptosis, inhibiting cell proliferation, migration, invasion, and angiogenesis, improving immune function and tumor microenvironment, and regulating related signaling pathways. Clinically, Xihuang pill, as an adjuvant drug in combination with chemotherapy, can prolong survival, enhance response rate, improve quality of life, regulate immune function, and alleviate chemotherapy-induced toxicities.

Infectious Mimics of Inflammatory Bowel Disease

Nicole C. Panarelli

Summary: Distinguishing inflammatory bowel disease (IBD) from its mimics is challenging for surgical pathologists due to overlapping inflammatory patterns produced by several gastrointestinal infections. Clinical tests may not always be available or reflect ongoing infections. Therefore, surgical pathologists should be knowledgeable about infectious mimics of IBD to generate accurate diagnoses and guide clinical follow-up. This review focuses on bacterial, fungal, and protozoal infections in the differential diagnosis of IBD.

Ursolic Acid Protects Sodium Dodecyl Sulfate-Induced Drosophila Ulcerative Colitis Model by Inhibiting the JNK Signaling

Tian Wei et al.

Summary: Ursolic acid (UA), a bioactive molecule found in fruits and vegetables, has therapeutic potential in ulcerative colitis (UC) induced by toxic chemicals. In this study, using a Drosophila UC model, the protective effect of UA on intestinal damage induced by sodium dodecyl sulfate (SDS) was investigated. The results showed that UA could protect against SDS-induced damage by reducing oxidative stress and restoring the activity of antioxidant enzymes. Additionally, UA inhibited the excessive activation of the JNK-dependent JAK/STAT signaling pathway. These findings suggest that UA has potential for the development of functional food or natural medicine for UC.

STING1 is essential for an RNA-virus triggered autophagy

Rui Zhang et al.

Summary: The study reveals that during foot-and-mouth disease virus (FMDV) infection, the integrated stress response in cells is initiated by the cellular pattern recognition receptor DDX58/RIG-I, which signals to the ER-anchored adaptor protein STING1. This activation leads to reticulophagy and degradation of STING1, which is essential for FMDV genome replication and viral protein expression.

The influence of cytokines on the complex pathology of ulcerative colitis

Hiroshi Nakase et al.

Summary: Ulcerative colitis (UC) is specifically affected by genetic alterations, environmental factors, microbiota, and mucosal immune dysregulation, leading to a lack of response in 30%-55% of patients to current therapies. The expression levels of cytokines associated with treatment depend on disease stage and patient characteristics, suggesting that future targeted therapies should focus on these differences and individual cytokine profiles.

Colorectal Cancer in Inflammatory Bowel Disease: Mechanisms and Management

Summary: Patients with inflammatory bowel disease (IBD) have an increased risk of developing colorectal cancer (CRC). Chronic inflammation plays a key role in the development of neoplastic progression, resulting in dysplastic precursor lesions. Colitis-associated CRC shares molecular similarities with sporadic CRC and the microbiome and host immune system may also play a role. However, further research is needed to understand the specific molecular differences and their role in inflammation-associated cancers.

ENO2 Promotes Colorectal Cancer Metastasis by Interacting with the LncRNA CYTOR and Activating YAP1-Induced EMT

Chunwei Lv et al.

Summary: This study found that upregulation of the glycolytic enzyme enolase 2 (ENO2) in colorectal cancer (CRC) samples was negatively associated with prognosis. Furthermore, the study showed that ENO2 promotes CRC cell migration and invasion through its interaction with the long noncoding RNA (lncRNA) CYTOR. CYTOR mediates the binding of ENO2 to large tumor suppressor 1 (LATS1) and competitively inhibits the phosphorylation of Yes-associated protein 1 (YAP1), ultimately triggering epithelial-mesenchymal transition (EMT).

Pioglitazone Attenuates the Effects of Peripheral Inflammation in a Human In Vitro Blood-Brain Barrier Model

Gustavo Henrique Oliveira da Rocha et al.

Summary: This study investigated the effects of plasma from inflammatory bowel disease (IBD) patients and tumor necrosis factor alpha (TNF alpha) on the blood-brain barrier (BBB), as well as the anti-inflammatory role of pioglitazone. The results showed that TNF alpha increased BBB permeability, while pioglitazone attenuated these effects. Plasma from IBD patients also increased BBB permeability, which was reversed by pioglitazone. These findings demonstrate the potential of pioglitazone in controlling peripheral BBB inflammation associated with inflammatory conditions.

Increased STING expression is a feature of intestinal inflammation in both mice with colitis and humans with inflammatory bowel disease. STING accumulation in intestinal myeloid cells can be triggered by dysbiosis and bacterial products, leading to a positive feedback loop that drives intestinal inflammation.
