Design and synthesis of new adamantyl derivatives as promising antiproliferative agents

Summary: A series of adamantyl carboxamide derivatives containing sulfonate or sulfonamide moiety were designed and evaluated for their antiproliferative activity against NCI-60 cancer cell lines panel. Compounds 1e and 1i exhibited potent and broad-spectrum inhibition against multiple cancer subtypes while showing safety towards normal cells.

Inhibitors of ecto-5'-NT can prevent tumor progression, lower adenosine concentrations, and impede cancer growth by preventing immune system evasion.

Synthesis and Biological Evaluation of Arylamide Sulphonate Derivatives as Ectonucleotide Pyrophosphatase/Phosphodiesterase-1 and-3 Inhibitors

Summary: Aberrant levels of ectonucleotide pyrophosphatase/phosphodiesterase-1 and -3 are associated with various disorders, and their activity can be inhibited by arylamide sulphonates. Selective inhibitors of ENPP1 showed promising results, with compounds 4f and 4q exhibiting sub-micromolar IC(50) values. Compounds 4t and 7d selectively inhibited the activity of ENPP3. The tested compounds showed significant inhibition of both isozymes, and molecular docking analysis revealed their interactions with influential amino acids.

Summary: A series of sulfonylurea derivatives possessing pyrrolo[2,3-b]pyridine core were synthesized and investigated as inhibitors of NPP1 and NPP3 isozymes. Compound 1c and 1h showed the strongest cytotoxicity against cancer cell lines MCF-7 and HT-29, respectively. These compounds exhibited promising interactions with important binding sites residues of NPP1 and NPP3, making them potential candidates for cancer and diabetes treatment.
