Elicitation for activation of the actinomycete genome's cryptic secondary metabolite gene clusters

Seham S. S. El-Hawary et al.

Summary: This review summarizes recent advances in the elicitation approaches used to activate cryptic secondary metabolite gene clusters in the actinomycete genome, highlighting the diversity of natural products obtained through various elicitation methods until June 2022, including co-cultivation with actinomycetes, non-actinomycete bacteria, fungi, cell-derived components, and/or algae. Chemical and molecular elicitation strategies such as transcription factor decoys, engineering regulatory genes, promoter replacement, global regulatory genes, and reporter-guided mutant selection were also discussed. This review serves as a valuable resource for researchers interested in this field, providing the latest studies and references.

Untargeted LC-MS/MS Metabolomics Study on the MCF-7 Cell Line in the Presence of Valproic Acid

Alan Ruben Estrada-Perez et al.

Summary: Research has found that valproic acid (VPA), a well-known histone deacetylase inhibitor (HDACi), can induce cell death in breast cancer cells by altering metabolic pathways. This includes metabolites that reduce cell proliferation and growth, interfere with energy sources, and increase reactive oxygen species levels. These findings suggest that VPA has different pharmacological mechanisms in killing cancer cells, and metabolomic approaches are a valuable source of information for understanding these mechanisms.

Identification of gingerenone A as a novel senolytic compound

Ruin Moaddel et al.

Summary: Senescent cells, which accumulate with aging, have been shown to contribute to age-associated diseases and organ dysfunction. In this study, ginger extract was found to selectively eliminate senescent cells without affecting normal cells. The major components of ginger extract, gingerenone A and 6-shogaol, showed promising senolytic properties and also suppressed the senescence-associated secretory phenotype (SASP). These findings suggest that gingerenone A may have therapeutic benefit in diseases characterized by senescent cell accumulation.

Compendium of specialized metabolite biosynthetic diversity encoded in bacterial genomes

Athina Gavriilidou et al.

Summary: By analyzing a large number of bacterial genomes and metagenome assembled genomes (MAGs), it was found that only about 3% of the potential natural products encoded in bacterial genomes have been characterized. The study also identified the genus level as an appropriate taxonomic rank for comparing the biosynthetic diversity of secondary metabolites, with Streptomyces bacteria encoding the largest diversity. Additionally, less-studied taxa such as Weeksellaceae, Myxococcaceae, Pleurocapsa, and Nostocaceae were found to have the potential to produce diverse secondary metabolites that require further investigation.

Antifungal Activity of Fibrate-Based Compounds and Substituted Pyrroles That Inhibit the Enzyme 3-Hydroxy-methyl-glutaryl-CoA Reductase of Candida glabrata (CgHMGR), Thus Decreasing Yeast Viability and Ergosterol Synthesis

Damian A. Madrigal-Aguilar et al.

Summary: Due to the emergence of multidrug-resistant strains of yeasts, discovering antifungal agents that target alternative molecular targets is urgently needed. This study evaluated the capacity of three different series of synthetic compounds to inhibit a specific enzyme in Candida glabrata, leading to the disruption of ergosterol synthesis and yeast viability. Two compounds, 1c and 5b, were identified as the best antifungal candidates among the compounds studied. They effectively inhibited the growth of fluconazole-resistant and fluconazole-susceptible C. glabrata strains. The compounds act by inhibiting the mevalonate synthesis pathway, resulting in a decrease in ergosterol concentration. The docking analysis suggested that their inhibitory effect may be mediated by their interaction with the enzyme's catalytic site. Further research and modifications to increase their specificity and potency are needed.

Volatile Compounds in Actinomycete Communities: A New Tool for Biosynthetic Gene Cluster Activation, Cooperative Growth Promotion, and Drug Discovery

Lorena Cuervo et al.

Summary: This study highlights the importance of discovering new therapeutic alternatives, especially natural compounds that can be used to treat multiresistant pathogens and emerging diseases. The research finds that volatile compounds (VOCs) have the ability to activate biosynthetic pathways of bioactive compounds in Actinomycetes, leading to increased antibiotic production and synthesis of new VOCs. The results demonstrate a novel strategy to exploit the metabolic potential of Actinomycetes and emphasize the significance of studying interactions between different microorganisms sharing the same ecological niche.

Review of Treatments for Oropharyngeal Fungal Infections in HIV/AIDS Patients

Alexandre Noel de Tilly et al.

Summary: This review summarizes the treatment options for oropharyngeal fungal infections in HIV/AIDS patients, including azoles, polyenes, and novel treatments. The studies showed increased reports of antifungal resistance, especially fluconazole resistance. Nystatin was recommended as a first-line therapy, but miconazole nitrate may be a better choice in resource-poor settings. Amphotericin B was an effective treatment option with notable side effects. Alternative treatments such as immunoglobulin antibodies and lemon grass showed promising antifungal effects.
