The ERAD system is restricted by elevated ceramides

Jiwon Hwang et al.

Summary: Misfolded proteins in the ER are removed through ERAD, a process involving membrane protein quality control. While ERAD is known to regulate lipid biosynthetic enzymes, the impact of lipid composition on ERAD is unknown. This study shows that specific ceramides in the ER membrane can affect ERAD function by impairing substrate extraction.

HERPUD1, a Member of the Endoplasmic Reticulum Protein Quality Control Mechanism, may be a Good Target for Suppressing Tumorigenesis in Breast Cancer Cells

Yalcin Erzurumlu et al.

Summary: This study evaluated the possibility of HERPUD1 as a potential therapeutic target for breast cancer. The results showed that suppressing HERPUD1 expression reduced cell cycle-related protein levels and decreased levels of EMT-related N-cadherin and angiogenesis marker vascular endothelial growth factor A in MCF-7 cells. Moreover, silencing of HERPUD1 significantly limited cell proliferation, migration, invasion, and colony formation of MCF-7 cells.

Inhibition of USP14 influences alphaherpesvirus proliferation by degrading viral VP16 protein via ER stress-triggered selective autophagy

Sheng-Li Ming et al.

Summary: Infection by alphaherpesvirus causes severe health consequences in various hosts. USP14, as the largest subfamily of deubiquitinating enzymes, plays a crucial role in regulating viral replication and degradation of viral proteins. Targeting USP14 could provide a potential therapeutic strategy for treating alphaherpesvirus-induced infectious diseases.

PCBP2 maintains antiviral signaling homeostasis by regulating cGAS enzymatic activity via antagonizing its condensation

Haiyan Gu et al.

Summary: cGAS is regulated by its interacting protein PCBP2, which controls its enzyme activity and affects cGAS-STING signaling pathway in antiviral response. Overexpression of PCBP2 reduces the antiviral signaling while loss of PCBP2 increases cGAS activity. PCBP2 decreases cGAS enzyme activity by antagonizing cGAS condensation, ensuring appropriate cGAMP production and balanced cGAS-STING signal transduction.

Corosolic acid sensitizes ferroptosis by upregulating HERPUD1 in liver cancer cells

Yingxiu Peng et al.

Summary: This study found that corosolic acid (CA) can sensitize liver cancer cells to ferroptosis by inhibiting HERPUD1, which reduces glutathione (GSH) synthesis. Further experiments also confirmed that CA inhibits tumor growth through HERPUD1. These findings suggest that CA is a potential treatment for liver cancer.

HERPUD1 promotes ovarian cancer cell survival by sustaining autophagy and inhibit apoptosis via PI3K/AKT/mTOR and p38 MAPK signaling pathways

Xin Nie et al.

Summary: HERPUD1 is highly expressed in ovarian cancer and positively correlated with the expression of Lewis y antigen. HERPUD1 promotes ovarian cancer by promoting cell proliferation, inhibiting apoptosis, affecting the cell cycle, promoting autophagy, and inhibiting multiple signaling pathways.

TRIM47 accelerates aerobic glycolysis and tumor progression through regulating ubiquitination of FBP1 in pancreatic cancer

Lei Li et al.

Summary: TRIM47 is upregulated in pancreatic cancer, promoting cell proliferation and aerobic glycolysis by directly binding to and ubiquitinating FBP1, leading to cancer progression. Targeting the TRIM47/FBP1 axis may provide a novel strategy to suppress the development of pancreatic cancer.

Ubiquitination and degradation of NF90 by Tim-3 inhibits antiviral innate immunity

Shuaijie Dou et al.

Summary: NF90 is a virus sensor that initiates antiviral immunity by triggering SG formation, but its regulation by the Tim-3 pathway results in degradation of NF90 and inhibition of antiviral response. Activation of Tim-3 during VSV infection recruits TRIM47 to degrade NF90, but targeted inactivation of Tim-3 enhances SG formation downstream of NF90 and protects against VSV challenge. These findings provide insight into the crosstalk between Tim-3 and NF90 in antiviral immunity.
