DNA tetrahedral scaffold-corbelled 3D DNAzyme walker for electrochemiluminescent aflatoxin B1 detection

Summary: The immobilization efficiency of surface-based DNA walker can be improved by fixing three-dimensional (3D) DNAzyme walker on the top of DNA tetrahedral scaffold. With Ferrocene (Fc) labeled at the ends of substrate strand, the electrochemiluminescence (ECL) signal of Ru(bpy)2(cpaphen)2 +/- can be effectively quenched, resulting in a signal-off state of the sensor. Upon the addition of aflatoxin B1 (AFB1), the 3D DNAzyme walker is activated and releases Fc, thus recovering the ECL signal. The high movement efficiency of the 3D DNAzyme walker enables ultrasensitive detection of AFB1 with a detection limit of 0.58 fg mL-1 in the range of 1.0 fg mL- 1-10 ng mL-1. Satisfactory results were also obtained when detecting AFB1 in corn and peanut samples, indicating its potential application in food safety analysis.

DNAzyme-driven tripedal DNA walker mediated signal-on and label-free for electrochemical detection of α-synuclein oligomers

Summary: In Parkinson's disease, the soluble alpha-synuclein oligomer is a reliable molecular biomarker. This study presents an innovative label-free and signal-on electrochemical apta-sensor for the quantification of alpha-syn oligomers. The tripedal DNA walkers triggered by alpha-syn oligomers outperformed common DNA walkers, leading to greater amplification efficiency. With the assistance of Mg2+-dependent DNAzyme, the tripedal DNA walkers could scissor DNA hairpins, generating enhanced current signals. The developed aptasensor demonstrated high sensitivity, anti-interference, and reproducibility, making it a promising tool for bioanalysis in complex biological environments.
