Effects of Sirolimus treatment on patients with β-Thalassemia: Lymphocyte immunophenotype and biological activity of memory CD4+ and CD8+ T cells

Matteo Zurlo et al.

Summary: Inhibitors of mTOR, such as Sirolimus, can enhance vaccine responses in the elderly. Testing Sirolimus and other geroprotective drugs might be a key strategy to improve the overall health resilience of aged populations. This study analyzed the activity of CD4(+) and CD8(+) T cells in beta-Thalassemia patients treated with Sirolimus and found that it positively impacted the biological activity and number of memory T cells.

Adjustment of Immunosuppressants to Facilitate Anti-COVID-19 Antibody Production after mRNA Vaccination in Liver Transplant Recipients

Wei-Chen Lee et al.

Summary: Liver transplant recipients were found to have low immunogenicity for anti-COVID-19 antibodies. Temporarily suspending the use of immunosuppressant drugs such as MMF or EVR during Moderna mRNA-1273 vaccination can facilitate antibody production. Factors such as temporary drug suspension and monotherapy were found to be favorable for generating a humoral response to the vaccine.

Evaluation of off-label rapamycin use to promote healthspan in 333 adults

Tammi L. Kaeberlein et al.

Summary: Rapamycin, an FDA-approved drug, has been shown in preclinical studies to extend lifespan and healthspan in various organisms. Some physicians are prescribing it off-label for preventative therapy, but there is limited data on its side effects or efficacy in this context. In this study, data was collected from adults who have used rapamycin off-label and compared to a group who have never used it, suggesting that rapamycin can be safely used in adults of normal health status.

mTOR inhibitors improve both humoral and cellular response to SARS-CoV-2 messenger RNA BNT16b2 vaccine in kidney transplant recipients

Giuseppe S. Netti et al.

Summary: mTOR inhibitors (mTOR-I) treatment is associated with a better immune response to COVID-19 vaccine in kidney transplant recipients (KTR), not only by increasing vaccine-induced antibodies but also by stimulating anti-SARS-CoV-2 T cell response.

Possible effects of sirolimus treatment on the long-term efficacy of COVID-19 vaccination in patients with β-thalassemia: A theoretical perspective Hypothesis and Theory

Matteo Zurlo et al.

Summary: The COVID-19 pandemic has been a challenge for global health, emphasizing the need for effective vaccines. Recent studies have shown that patients with beta-thalassemia have a high mortality rate from COVID-19, likely due to their immune deficiencies. Immunosuppression may also impact the effectiveness of COVID-19 vaccines, suggesting the use of immunomodulatory molecules as a potential solution.

Eli S. Rosenberg et al.

Summary: The effectiveness of the BNT162b2, mRNA-1273, and Ad26.COV2.S vaccines in New York State from May to August 2021 was assessed. Initial protection was high, but there was a modest decline in effectiveness after the surge of the delta variant, especially among recipients aged 65 and older. However, protection against hospitalization remained high.

Summary: This study evaluated the humoral response to the Comirnaty vaccine in patients with Thalassemia Major (TM) and compared it with healthy blood donors (HBD). The results showed a similar serological response to the vaccine in both groups. Interestingly, asplenic patients had higher antibody titers, which may be related to splenectomy.

Immunomodulatory and Antiaging Mechanisms of Resveratrol, Rapamycin, and Metformin: Focus on mTOR and AMPK Signaling Networks

Vincenzo Sorrenti et al.

Summary: Aging is a result of the dysregulation of multiple molecular pathways, with mTOR and AMPK signaling playing a role in cellular senescence. Poor nutritional balance is a driving factor in immunosenescence progression, and nutraceutical and pharmacological interventions can help maintain optimal biological response. Compounds like resveratrol, rapamycin, and metformin have shown positive effects in healthy aging.

Summary: Patients with beta-thalassemia have a 5-fold increase in age-standardized lethality due to SARS-CoV-2 infection, making them a high-risk population. Vaccination is crucial in reducing mortality and morbidity for these frail patients. Limited data is available on the response of beta-thalassemia patients immunized with anti-SARS-CoV-2 mRNA vaccines.
