Bile Acid Conjugation on Solid Nanoparticles Enhances ASBT-Mediated Endocytosis and Chylomicron Pathway but Weakens the Transcytosis by Inducing Transport Flow in a Cellular Negative Feedback Loop

Summary: Bile acid-modified nanoparticles can improve oral bioavailability of poorly permeable drugs by interacting with bile acid transporters. This study investigates the transport of glycocholic acid (GCA)-conjugated polystyrene nanoparticles (GCPNs) in Caco-2 cell models and identifies a new pathway correlated with both apical sodium-dependent bile acid transporter (ASBT) and chylomicron pathways. The study also reveals that the higher uptake of GCPNs does not result in higher transcytosis compared to unmodified nanoparticles (CPNs). Pharmacological and genomics analysis indicate that GCA conjugation alters endocytosis mechanisms and downregulates cellular response, leading to higher cellular retention of GCPNs.

Dual crosslinking of folic acid-modified pectin nanoparticles for enhanced oral insulin delivery

Summary: Pectin-based drug delivery systems have great potential for oral insulin delivery. In this study, we developed folic acid (FA)-modified pectin nanoparticles (INS/DFAN) as insulin delivery vehicles through a dual-crosslinking method. INS/DFAN effectively prevented premature insulin release and enhanced insulin transepithelial transport via the FA carrier-mediated pathway. In vivo studies showed that orally-administered INS/DFAN significantly reduced blood glucose levels and improved insulin bioavailability in type I diabetic rats compared to ionic-crosslinked nanoparticles (INS/FAN). The combination of dual crosslinking and FA modification is an effective strategy for enhanced oral insulin delivery using pectin nano-vehicles.

Exploring protein stabilized multiple emulsion with permeation enhancer for oral delivery of insulin

Summary: In this study, a novel oral insulin microemulsion was developed with piperine as a permeation enhancer and albumin as a stabilizer. The microemulsion showed small droplet size and good stability, maintaining INS stability in simulated gastrointestinal fluids and showing improved uptake and permeation compared to free INS. Animal studies demonstrated a significantly higher hypoglycemic effect and pharmacokinetic studies revealed higher AUC for the microemulsion with albumin as a stabilizer, suggesting its potential for oral delivery of bio-macromolecules.
