The Zoonotic Potential of Chronic Wasting Disease-A Review

Novel Prion Strain as Cause of Chronic Wasting Disease in a Moose, Finland

Summary: Our previous studies found that different strains of prions cause chronic wasting disease (CWD) in Norway and North America cervids, and they respond differently to amino acid variation at residue 226 of the prion protein. This study investigated the properties of newly emergent CWD in moose in Finland using gene-targeted mice transmissions. The findings showed that the strain properties of Finland moose CWD prions are distinct from those previously characterized in Norway CWD, indicating a diverse portfolio of emergent strains in Nordic countries.

Chronic wasting disease in Norway-A survey of prion protein gene variation among cervids

Summary: The susceptibility of cervids to Chronic Wasting Disease (CWD) in Norway is modulated by variations in the prion protein gene (PRNP), with different species having different levels of susceptibility. The PRNP genetic data indicate considerable susceptibility to CWD among Norwegian cervids, and suggest that PRNP homozygosity may be a risk factor for the atypical CWD observed in moose. The CWD isolates found in Norwegian cervids differ from those previously found in Canada and USA, indicating potential strain dynamics related to PRNP variability.

Summary: This study evaluated the amplification of PrPTSE from human sCJD brain samples in different substrates using PMCA, revealing the potential of bank vole PrP substrate in amplifying all sCJD subtypes. In contrast, PMCA in human PrP substrates led to molecular shifts in PrPTSE, with increased permissiveness of V129 PrP substrate to sCJD prion amplification. The combination of PMCA sensitivities and PrPTSE electrophoretic profiles confirmed the classification of 4 distinct major sCJD prion strains. Additionally, the sensitivity required to detect VV2 sCJD prions in cerebrospinal fluid was achieved.
