Upper respiratory tract mucosal immunity for SARS-CoV-2 vaccines

Summary: SARS-CoV-2 vaccination reduces morbidity and mortality, but has limited impact on viral transmission and waning immunity. Immune responses in respiratory tract mucosal tissues are essential for infection control, but current vaccines cannot effectively target the upper airways, leading to breakthrough infections and limited duration of protection. To address these issues, a strategy is proposed to enhance vaccine efficacy and durability by inducing robust memory responses in the upper respiratory tract mucosa.

Studies have shown that SARS-CoV-2 proteins Nsp6 and Orf8 inhibit the response to type I interferon at different stages, and Orf6 also inhibits the activation of IRF3 and TNF-alpha-induced NF-kappa B activation.

Summary: The third outbreak of coronavirus (CoV) infection caused by SARS-CoV-2 has become a global pandemic, with the virus entering host cells primarily through the interaction between viral proteins and the ACE2 cell surface protein. Strategies to target virus entry into cells include direct targeting of viral glycoproteins, viral receptors on the cell surface, and the use of soluble ACE2 to neutralize the virus. Fusion of sACE2 to anti-CD16 to produce a bi-specific molecule could be a promising anti-viral strategy.
