Highly reproducible and sensitive electrochemical biosensor for Chlamydia trachomatis detection based on duplex-specific nuclease-assisted target-responsive DNA hydrogels and bovine serum albumin carrier platform

Lingjun Cheng et al.

Summary: In this study, a highly reproducible and sensitive electrochemical biosensor was developed for the detection of Chlamydia trachomatis (CT). The biosensor is based on duplex-specific nuclease (DSN)-assisted target-responsive DNA hydrogels and a bovine serum albumin (BSA) carrier platform. The biosensor can reliably detect CT 16S rRNA. The signal conversion function of the DNA hydrogel and the optimization by BSA provide high reproducibility to the biosensor.

Sensitive Electrochemiluminescence Biosensor Based on the TargetTrigger Difference of the Electrostatic Interaction between an ECLReporter and the Electrode Surface

Yilei Lu et al.

Summary: By utilizing the discrepancy of electrostatic interaction between negatively charged signal molecules and long and short DNA strands on the modified electrode surface, an electrochemiluminescence (ECL) biosensor for HPV 16 DNA detection has been developed. This innovative methodology offers sensitive detection of HPV concentration by controlling electrostatic interaction and steric hindrance.

Dual-enzyme cascade amplification electrochemical biosensor for human papillomavirus based on DNA nanoflower structure

Hongzhang He et al.

Summary: The lack of large-scale HPV DNA screening is a major driver of high cervical cancer incidence and mortality in economically undeveloped areas. In this study, DNA nanoflowers were synthesized as a carrier for HPV DNA detection, offering a sensitive, rapid, and low-cost approach. The proposed method has the potential for large-scale HPV DNA screening in economically underdeveloped areas, as well as serving as a blueprint for detecting other DNA of interest.

Highly Reproducible and Sensitive Electrochemiluminescence Biosensors for HPV Detection Based on Bovine Serum Albumin Carrier Platforms and Hyperbranched Rolling Circle Amplification

Yinghao He et al.

Summary: In this study, a porous BSA layer was modified on the electrode surface, improving the distribution and orientation of self-assembled ssDNA probes, coupled with hyperbranched rolling circle amplification to develop a high-reproducibility-and-sensitivity ECL biosensor for human papillomavirus 16 E6 and E7 oncogene detection. The sensor showed a linear relationship between signal and target concentration, with a detection limit of 7.6 fM and improved reproducibility after BSA modification. The proposed ECL strategy offers a new method for constructing high-reproducibility-and-sensitivity ECL biosensors.

Modeling Effects of Surface Properties and Probe Density for Nanoscale Biosensor Design: A Case Study of DNA Hybridization near Surfaces

Timothy Cholko et al.

Summary: Electrochemical biosensors have versatile applications due to their robust nature and the ability to design highly sensitive sensors through optimizing the arrangement and properties of immobilized probes. The distribution of probes on the sensor surface may affect the efficiency of target-probe association, while controlling the interprobe spacing can enhance hybridization efficiency. Simulation studies provide insights into the dynamics of hybridization processes, aiding in the design of efficient biosensors and understanding experimental trends at different probe densities.

Double Hairpin DNAs Recognition Induced a Novel Cascade Amplification for Highly Specific and Ultrasensitive Electrochemiluminescence Detection of DNA

Xiaoli Zhang et al.

Summary: A novel DNA cascade amplification method was developed as a signal amplifier for constructing a highly specific and ultrasensitive electrochemiluminescence (ECL) biosensor for detecting HIV DNA fragments. The RCA products anchored on the Pt-modified GCE via Pt-N bond allows for capturing Ru-labeled ssDNA as the ECL signal tag, achieving highly specific and ultrasensitive detection of the target.
