Glucose transporter 4: Insulin response mastermind, glycolysis catalyst and treatment direction for cancer progression

Yu-Chan Chang et al.

Summary: The glucose transporter family (GLUT) consists of fourteen members and is responsible for glucose homeostasis and transport. Different GLUTs have unique kinetics and distribution, playing important roles in human physiology. GLUT4 mediates insulin-dependent glucose uptake and dysfunction of GLUT4 is associated with insulin resistance, chronic diseases, inflammation, and cancer.

Helichrysetin inhibits gastric cancer growth by targeting c-Myc/PDHK1 axis-mediated energy metabolism reprogramming

Ping Wang et al.

Summary: Helichrysetin (HEL) isolated from Alpinia katsumadai Hayata inhibits gastric cancer growth by targeting the mTOR/p70S6K/c-Myc/PDHK1 mediated energy metabolism reprogramming in cancer cells. It significantly inhibits cell viability, glycolysis, and lactate production, and modulates mitochondrial oxidative phosphorylation, ROS accumulation, and ATP production. Additionally, HEL inhibits the overactivated mTOR/p70S6K pathway both in vitro and in vivo.

Isoliquiritigenin Inhibits Gastric Cancer Stemness, Modulates Tumor Microenvironment, and Suppresses Tumor Growth through Glucose-Regulated Protein 78 Downregulation

Chien-Hsing Lee et al.

Summary: Chemotherapy is commonly used for gastric cancer treatment, but its efficacy is limited. This study showed that glucose-regulated protein 78 (GRP78) plays a crucial role in gastric cancer stemness, and isoliquiritigenin (ISL) can inhibit GRP78, thereby suppressing gastric cancer stem cell-like characteristics and tumor growth, as well as promoting chemosensitivity.

Hypoxia-induced FOXO4/LDHA axis modulates gastric cancer cell glycolysis and progression

Xiao-Hong Wang et al.

Summary: FOXO4 downregulation in gastric cancer is associated with poor prognosis. Restoring FOXO4 expression decreases glycolysis rate in GC cells, while silencing it increases glycolysis rate. FOXO4 is a transcriptional target of HIF-1 alpha and plays a key role in regulating glycolysis in GC.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: The study revealed that ISL1 enhances glycolysis and tumorigenesis in gastric cancer by transcriptionally regulating GLUT4. High levels of ISL1 expression were associated with shorter survival of GC patients, while ISL1 knockdown inhibited cell proliferation and impaired glycolysis.
